Cancer has become the leading cause of death in human world. Drug-loaded liposomes will decrease the toxic side effect on patients. However, the application is limited for its low stability and poor drug release properties. Hybrid lipid with hydrophobic carbochain and hydrophilic siloxane could form into liposomal nanobybrid cerasomes based on sol-gel method and self-assembling. Drug-loaded cerasomes have a perfect performance in stability and drug release properties. However, defined as a novel drug carrier, influence of chemical structure of hybrid lipid on the drug release properties is not clear enough.In order to study influence of the chemical structure of hybrid lipid on the drug release properties, in this paper, blank cerasomes were fabricated from four hybrid lipids with four chemical structures. The diameters of these nanoparticles were about 200nm, with negative electricity, strong stability, favorable dispersivity and nontoxicity. Afterwards, paclitaxel (PTX) and doxorubicin (DOX) as the model drug were loaded into four kinds of cerasomes based on Bangham's method. Drug release properties of four cerasomes and entrapment efficiency were studied and the number of hydrophobic carbochain and hydrophilic siloxane having the effect on the drug release speed was analized. Based on the results, the entrapment efficiency of PTX-loaded cerasomes is increasing with proportion of hydrophobic carbochain rising and the entrapment efficiency of DOX-loaded cerasomes is increasing with proportion of hydrophilic siloxane rising. Drug release speed of both of the drug-loaded cerasomes is decreasing with the number of hydrophilic siloxane rising, for the condensation of Si-O-Si matrix. Cytotoxicity essays showed that all the drug-loaded cerasomes have controlled-release toxicity on the Hela cell line.In conclusion, four of different structures of hybrid lipids were applied in loading PTX and DOX. The effect of chemical structure of hybrid lipids on the entrapment efficiency and drug release properties was studied and the disciplines were concluded in this essay. This is significance for drug-loaded cerasomes with different drug release properties designed in future.
