【Objective】To detect the levels of the mRNA expression of tumor necrosis factor like ligand 1A(TL1A) and Indoleamine-2,3-dioxygenase(IDO) in the peripheral blood monocytes(PBMCs) of the patients with rheumatoid arthritis(RA) and explore their potential role in the pathogenesis of RA.【Methods】Real time fluorescence quantitative RT-PCR was performed to measure TL1A mRNA and IDO mRNA expression in PBMCs of 60 cases with RA,60 other disease controls and 60 healthy controls. Andβ-actin was used as internal control.The comparison of gene expression level was applied with the formula of delta delta Ct .【Results】The relative level of TL1A mRNA expression of RA patients 3.65±0.63 was statistically higher than that of disease controls 1.08±0.42 (t=7.32, P <0.01) and that of healthy controls 1.04±0.40 (t=9.43,P<0.01) .The relative level of IDO mRNA expression of RA patients 2.63±0.51 was also significantly higher than that of disease controls 1.13±0.45(t=6.28, P<0.01) and that of healthy controls 1.05±0.52(t=8.82, P<0.01).The RA patients in active phase had significantly higher level of TL1A expression 3.85±0.43 and IDO expression 2.91±0.38 than that of RA patients in non-active phase TL1A 2.39±0.62(t=8.54, P<0.01) , IDO 1.81±0.54(t=6.98, P<0.01), respectively . Among the RA patients,no statistically significant difference was observed between the groups of the males TL1A 3.35±0.68 and the females TL1A 3.45±0.79(t=0.395,P>0.05);the males IDO 2.42±0.56 and the females IDO 2.51±0.64(t=0.475, P>0.05).There was statistical difference in the expression of TL1A between RF(+) RA group and RF(-) RA group(t=5.85, P<0.01).But no statistically significant difference was found in the expression of IDO between RF(+) RA group and RF(-) RA group(t=0.425,P>0.05).【Conclusion】Compared with the healthy controls and the disease controls,the level of TL1A mRNA expression and IDO mRNA expression of PBMCs are both significantly augmented in patients wih RA,especially in the active phase of RA.There is no significant difference in the expression of TL1A and IDO between genders.The TL1A and IDO may be involved in the process of inflammation and pathogenesis of RA.
