Objective To investigate the effections of Sema3A which is one of semaphrins family on the expressions of B7-H3 on murine bone-marrow-derived dendritic cells(DCs).Method DCs from murine bone-marrow-derived were prepared,including imDCs and mDCs.On the eighth day,DCs were sorted and detected CD11C,CD40, CD80,CD86,B7-H3 expression.The imDCs and mDCs were separately divided into two groups,one is the control group and another is intervention group.The intervention group treated with different concentrations of Sema-3A-Ab.72 hours after intervention,the expressions of B7-H3 on imDCs were examed by flow-cytometry.Results Collected by flowcytometry of DC,CD11c+ cells reached 82.18%.The expression of B7-H3 on mature DC was (7.22±0.44)% on 8 day less than (19.68±0.66)% on 11 day.Sema-3A-Ab down-regulated the expression of B7-H3 on mature DC derived from murine bone-marrow,intervention group(10.15±0.91)%,control group(19.68±0.66)%,(p<0.05)Conclusion The expression of B7-H3 on mature DC was rising with time. Sema-3A-Ab down-regulated the expression of B7-H3 on mature DC.Sema3A can increase the expression of B7-H3 on mature DC. The current study was no significant difference between the intervention group of different concentration.
