Objective: To investigate the expressions of S-phase kinase-associated protein 2(Skp2)and p27 in the hippocampus after reperfusion following forebrain ischemia in rats.Methods: 90 SD wistar rats were randomly divided into two groups : control group(n=30),ischemia-reperfusion group(n=60). The rats of ischemia-reperfusion group were induced by pulsinelli-4-vessel occlusion method,the carotid clasps were removed after 30min of 4-vessel occlusion and restoration of carotid blood flow. The rats of control group received sham operation. We investigated the change of neurons and astrocytes in hippocampus by immunohistochemistry 4 and 7 days after reperfusion. The expression of cyclinB1 mRNA was detected by quantitive real time PCR. Expressions of skp2 and p27 were measured by western blot at the same time.Results: Numbers of neurons in rat hippocampus decreased, while astrocytes increased significantly after the ischemia-reperfusion injury. Astrocytes activated obviously in rat brain after reperfusion. The expression of cyclinB1 mRNA in ischemia-reperfusion group was significantly higher than those in control group (P<0.05). Expressions of skp2 increased markedly after 4 and 7 days of reperfusion while p27 decreased after 4 and 7 days of reperfusion compared with rats in control group (P<0.05). Conclusion: Skp2 was increased in hippocampus after reperfusion following forebrain ischemia in rats,and such an event may be associated with neuronal apoptosis and glail proliferation after cerebral ischemia-reperfusion injury.
