| Objective: To observe the effect of chitooligosaccharides (COS) and saliva on the adhesive and detachment of Streptococcus mutans.Methods: S.mutans biofilm was formed on saliva-coated and uncoated glass slices, then exposed to COS with different concentrations for 30 minutes. Live/Dead fluorescent staining and confocal laser scanning microscopy technique were combined to observe the bacterial density, the data were analysis of variance.Results: The bacterial counts on saliva-coated and uncoated groups were decreased with the increasing of COS concentration ,significant difference was observed compared with the blank control group(P<0.05). The bacterial density on saliva-coated glass slices was more than uncoated glass slices after treated with the same concentration of COS, but there was no significantly difference in statistics(P>0.05).Conclusion: This study revealed that chitooligosaccharides was an antibacterial agent, it can be detaching biofilm effectively. The adhesion of S.mutans may be modulated by saliva and chitooligosaccharides . Objective: To study on detachment effect of chitooligosaccharides(COS) with different molecular weights on Streptococcus mutans biofilm.Methods: S.mutans biofilm was formed on saliva-coated glass in three six-well plates, then exposed to 2.5,5,10,20,40g/l COS solution with different molecular weights (2000,3000,5000) for 30min and 1h. Live/Dead fluorescent staining and confocal laser scanning microscopy technique were combined to observe the bacterial density.Results: The results indicats that bacterial density treated with three different molecular weights of COS solution all reduced significantly(P<0.05). When the same molecular weight COS,with the increase of COS concentration and treatment time,bacterial density decreased.When with the same treatment time and COS concentration,the results from strong to weak in promoting biofilm off were 5000,3000,2000,but there was no significant difference(P>0.05).Conclusion: The result demonstrate that molecular weight of chitooligosaccharides is a factor of detachment of S.mutans biofilm. |
