Object To observe whether Cdk1 expression and activity in HepG2 cells affects the cell apoptosis.Methods: The Cdk1 plasmid and Cdk1siRNA were constructed and transfected into hepatoma HepG2 cells. At the same time, the cells were treated by roscivintie, a Cdk1 inhibitor. After irradiation with UV light, Cdk1 protein expression was detected by Western Blot assay; The kinase activity was analysised by gama-32P labeled kinase assay; and cell apoptosis was detected by using HO 33342 (Hoechst 33342) staining.Results: Cdk1 plasmid could be expressed very well in HepG2 cells, and Cdk1 siRNA could efficiently silent the Cdk1 expression in HepG2 cells, and roscovintine removed the Cdk1 activity.The apoptosis rate is highest in the cells treated with roscovintine (p<0.01, vs to the cells transfected with Cdk1 and Cdk1siRNA), and is significantly high in the cells transfected with Cdk1, compared to the Cdk1 siRNA transfected cells (P<0.01) and control cells (EV cells) (P<0.05).Conclusions: Over expression of CDK1 or inhibition of its activity in HepG2 cells increases cell apoptosis and may play an important role in the therapy of liver cancer.
