Objective:â‘ To investigate the effect of the Annular Puncture and Aspiration disc degeneration model;â‘¡To investigate the effect that LINK-N/RADA-16 carry MSCs to repair the degenerate intervertebral disc.Methods:Rabbit bMSCs were cultured in vitro and the Self-assembled peptide nanofiber material LINK-N/RADA-16 was compounded; the treatment to each intervertebral disc segment is in the following: L2/3-only Annular Puncture and Aspiration; L3/4-after puncture and aspiration RADA16 +MSCs was injected; L4/5-after puncture and aspiration LINK-N/RADA-16+MSCs was injected, L1/2 segment was as control.Before the treatment and three weeks later MRI was obtained, and then the rabbits were sacrificed for histological analysis and immunohistochemistry of the intervertebral disc tissue.Results:â‘ MRI results showed that the height the L2/3 was lower and the image singal was decreased in the T2W, compared with that the degeneration of the L3/4 segment was mild while the L4/5 segment was near to the normal control group.â‘¡The histology and immunohistochemistry results showed that nucleus pulposus cells was in a small number in the L2/3 segment and fibrous cartilage degeneration occured, at the same time, the level of collagenâ…¡was lower compared with the control group; in the L3/4 segment these changes were mild and the L4/5 segment was near to the control group.Conclusion:â‘ The method of punture and aspiration can effectively cause animal disc degeneration;â‘¡Implantation of bMSCs carried by LINK-N/RADA-16 can effectively inhabit the disc degeneration, so this material can be used as a good scaffolds of the disc tissue engineering.
