Objective:Investigated the effect and potential mechanism of combined plumbagin and docetaxel on apoptosis and growth inhibition of prostate cancer cell in vitro.Methods:Prostate cancer cells PC3 were used,which were randomized into four groups.One used as controls group.(C group), the others were treated with plumbagin(P group,5μM) ,docetaxel(D group,5nM) and their combination individually(P+D).After cultivation for 24~48hrs, the growth inhibition rates of PC-3 cells were studied by MTT colorimetry.Cellular apoptosis was detected by acridine orange-ethidium bromide fluorescent staining and Annexin V-FITC and propidium iodide staining flow cytometry(FCM).The mRNA and protein levels of NF-κB,Bcl-2,Bax and Vegf were determined by RT-PCR and western blotting.Results:Each intervention group could inhibit the growth of PC-3 in a dose- and time-dependent manner.The proliferation rates of D group,P group,D+P group in 24h were 19.11%,25.23%,46.53%, respectively. The inhibiting rate in D+P group was significantly higher than other groups(P<0.01 and P<0.05). The apotosis rates of D group,P group,D+P group in 24h were 5.1%, 20.5%, 29.4% and 51.2%.Theapotosis rate in D+P group was significantly higher than others(P<0.05).Partial cells presented the characteristic morphological changes of apotosis. The mRNA and protein levels of NF-κB,Bcl-2, and Vegf in D+P group were significantly reduced than others, however the mRNA and protein level of Bax was up-regulated.Conclusion:Plumbagin plus docetaxel could significantly inhibit the viability of PC-3 cells by suppressing proliferation and induing apotosis and its mechanisms might be invovlved in the down-regulation of NF-κB,Bcl-2 and Vegf,up-regulation of Bax gene.
