Objective To study the effect of global cerebral ischemia-reperfusion to the tight junction protein occluding in rat brain.Methods Thirty male Sprague–Dawley Rats were randomly divided into sham operation group, ischemia 30 min of reperfusion 3h, 12h, 24h, 72h .There are 5 groups and 6 rats in each. Sham operation group, only bilateral vertebral artery electro cauterize, ischemia-reperfusion group electro cauterize bilateral vertebral artery occlusion of bilateral common carotid artery for 30 minutes, and then kill the rats at each time point, and then remove the brain tissue. Protein and mRNA are extracted from the brain. Moreover, mRNA level of occludin and the expression level of occludin were measured by using Real-time PCR and Western blotting.Results Compared with the sham group, ischemia reperfusion group Occludin protein levels and mRNA were decreased(P<0.05), a slight decrease after reperfusion 3h, 12h significantly decreased, 24h to a minimum, 72h began to increase, but still lower than the sham operation group level.Conclusion cerebral ischemia-reperfusion expression of tight junction protein occluding decreased,and with the reperfusion time change.
