Objective. The aim of this study is to detect the expression of miR-146b-5p in pancreatic carcinoma cell lines and investigate the relationship between miR-146b-5p and the invasion and migration of pancreatic carcinoma cells. In addition it may provide new ideas for the study of development of pancreatic cancer.Methods1,The expression of miR-146b-5p in 6 pancreatic carcinoma cell lines was examined by quantitative Real-time PCR(qRT-PCR)。2,When MIA PaCa-2 cells have transfected with miR-146b-5p mimics ,the expression of miR-146b-5p in them was examined by qRT-PCR. Meanwhile, we investigated the ability of invasion and migration in MIA PaCa-2 cells by scratch migration assay and transwell invasion assay.ResultsCompared with normal pancreatic tissue, qRT-PCR showed that miR-146b-5p was poorly-expressed in six pancreatic carcinoma cell lines, especially in MIA PaCa-2. The expression level of miR-146b-5p was raised after transfected with miR-146b-5p mimics in MIA PaCa-2 cells, meanwhile the ability of invasion and migration was down-regulated.ConclusionsMiR-146b-5p was poorly-expressed in these pancreatic carcinoma cells. When miR-146b-5p has up-regulated in MIA PaCa-2, it could inhibit pancreatic carcinoma cell invasion and migration .,so that it is expected to become a new target mechanism.
