Objective: The cardiac allograft rejection is the main influential factor of the survival time of the patients. The patients often need long-term use immunosuppression drugs to combat rejection. The acute rejection after the transplantation is mainly caused by T cell-mediated cellullar immunologic response of the receptor. Therefore, the survival time of the patients could be prolonged by restraining this immune response. Our recent research has shown that BisⅧcould enhance the expression of CTIL-4 and inhibit PI-3K/PKB of activated T cells in cardiac allograft rejection. Based on the consequence before, we establish the rat cervical heterotopic heart transplantation model to discuss the suppression effect of BisⅧon cardiac allograft rejection in rats.Methods: The model of rat cervical heterotopic heart transplantation was set up, forty five rats with cervical heterotopic heart graft were divided into 3 groups randomly: group A were fed with saline 1ml per day, group B were fed with CsA 5 mg/kg per day, group C were fed with BisⅧ1mg/kg per day. Transplanted heart beating conditions were observed every day and grafts survival days were recorded, hepatic and renal function were detected on the third day after the operation.Results: In group B(16.4±1.6d) and C(17.2±1.8d), the survival time of the grafts was prolonged significantly as compared with group A(7.5±1.5d) (P<0.05) .There is no statistics difference of the survival time of the grafts between group B and group C (P >0.05). The hepatic and renal function of group B were worse than group A and C (P<0.05). The alanine aminotransferase(ALT), aspartate aminotransferase(AST) and blood urea nitrogen(BUN) is worse than group A (P<0.05). There is no statistics difference of creatinine(Cr) between group A and C (P>0.05).Conclusions: BisⅧcan suppress cardiac allograft rejection and has less influence on the hepatic and renal function than CsA.
