Abstract/Summary:
Objective: Oral Cavity Cancer is the sixth leading cancer worldwide. Oral Squamous Cell Carcinoma(OSCC) is the most common oral cavity cancer which accounts for more than 90% of total oral malignant tumors. It has a high incidence of invasion and metastasis, that is the leading cause of death. Ras/ Raf / MEK / ERK signal transduction pathway is frequently activated which always involved in invasion and metastasis in malignant tumors. In this experiment we study the expression of PTPIP51,p-Raf-1and ERK1/2 in OSCC and investigate their influence on invasion and metastasis of OSCC.Methods: The expression of PTPIP51 and p-Raf-1 was detected by immunofluorescence and confocal laser scanning microscope in 32 OSCC specimens and their remote normal mucosa specimen.The PTPIP51-positive cells of the carcinoma and the surrounding stroma were identified on the basesis of specific morphological feature by means of HE staining and immunohistochemistry (SP method) .Then total RNA were extracted from these specimens, Detection of PTPIP51 mRNA and ERK1/2 mRNA come done by Real-time PCR.Results: PTPIP51 and p-Raf-1 proteins were coexpression in malignant proliferating cells,immune cells and surrounding endothelial cells of vessels in OSCC. PTPIP51 mRNA and ERK1/2 mRNA in tumor specimens were significantly higher than their matched normal mucosa specimens. PTPIP51 mRNA and ERK1/2 mRNA have positive correlation (r=0.641, p<0.05). The mRNA of PTPIP51 and ERK1/2 in OSCC was higher in group with lymph node metastasis than that in group without lymph node metastasis (p<0.05). statistical significance Conclusion: PTPIP51 and Raf-1 may play a key role in the pathogenesis and development of oral squamous cell carcinoma via Ras/Raf/MEK/ERKpathway(extracellullar signal-regulated kinase pathway), and they are closely related to invasion and metastasis of OSCC. The expressions of PTPIP51 an Raf-1 were correlated with TNM staging and the lymph node metastasis,while non-correlated with age,gender and the pathological gradeing.
