BackgroundsPreeclampsia is a common obstetric diseases, is idiopathic hypertension during pregnancy may be associated with multiple organ damage. Preeclampsia and maternal mortality rate was 4.69%, and perinatal mortality in preeclampsia and eclampsia is as high as 33.9% o~58.6%.The pathogenesis of preeclampsia is unclear, since Starkt oxidative stress was first proposed in the early development of potential pathological role, more and more scholars begin to pay attention in oxidative stress and the pathogenesis of preeclampsia. Now that the incidence of preeclampsia and origin of the whole body oxidation-antioxidant imbalance, plasma inflammatory factors involved in endothelial injury, and further lead to systemic vascular endothelial dysfunction, vasoconstriction, end-organ hypoperfusion, increased vascular permeability, which The clinical symptoms caused by preeclampsia.Oxidative stress and endothelial dysfunction in a variety of diseases rela ted to play a key role. Preeclampsia and the body reactive oxygen species (ROS) and loss of balance between antioxidant systems, oxidative stress in the body.In normal pregnancy, pregnant women can also be seen free radical product, but because of pregnancy, a corresponding increase in antioxidant capacity between the two still in the balance, would not constitute oxidative stress. In recent years, found that proteins, peptides, amino acids and related oxides of various free radicals are quite sensitive. In preeclampsia, an increase in oxidation products (lipid peroxides, protein oxidation products, nucleic acid), reduced antioxidant capacity, leading to preeclampsia showed oxidative stress status in vivo.Advanced oxidation protein products (AOPP) is a novel protein markers of oxidative stress is oxidative stress caused by oxidation of the body formed by a variety of protein end products in general.By the Witko-Sarsat in 1996 first by patients with chronic renal failure in the isolated, contain a double-cross-linked protein tyrosine products, the main component of serum albumin by free radicals and reactive oxygen system (mainly neutrophil myeloperoxidase produced chlorinated oxides) after oxidation product. Recent studies found, AOPP levels reflect protein oxidation sensitive and reliable indicators, the main component of serum albumin by free radicals and reactive oxygen system (mainly neutrophil myeloperoxidase produced chlorinated oxides) oxideAfter the product.AOPP in vivo experiments can lead to endothelial cell damage, increased oxidative stress level. In diabetes, uremia, atherosclerosis, chronic enteritis and other diseases AOPPs average increase of water, which may be associated with disease formation.Kaneda and other studies have shown that AOPP is a reliable coronary artery disease risk factors. Hideaki Kaneda and other studies have shown that, like chronic coronary artery disease and uremic plasma protein oxidation. Patients with chronic coronary artery disease and uremic plasma AOPP levels also increased in the plasma of patients, and AOPP levels in chronic coronary artery disease related to the occurrence and severity.Oxidative stress may be relevant in the pathology of chronic coronary artery disease.AOPP as new markers of oxidative stress in preeclampsia and the role of research in the less, the paper by observing 30 patients with preeclampsia (including 15 cases of mild preeclampsia, severe preeclampsia 15 cases) and 20 normal Pregnancy, maternal plasma, umbilical cord blood of advanced oxidation protein products (Advanced oxidation protein products, AOPP) differences in expression levels; plasma malondialdehyde oxidation indicators (malondialdehyde, MDA), superoxide dismutase (superoxide dismutase, SOD) expression levels; plasma AOPP and cord blood AOPP, plasma MDA, serum SOD correlation; of AOPP in patients with preeclampsia and the relationship between oxidative stress.Observation By observing the advanced oxidation protein products (AOPP) in patients with preeclampsia and normal pregnant women and cord blood plasma differences in expression and plasma AOPP and cord blood AOPP, MDA, SOD correlation of AOPP in the pathogenesis of preeclampsia The relevance of AOPP in exploring the pathogenesis of preeclampsia and the role of oxidative stress for the further study of the pathogenesis of preeclampsia to provide clues to the molecular pathway.Research Methods1. Using spectrophotometric detection of preeclampsia and normal pregnancy, maternal plasma AOPP, the expression of cord blood levels of AOPP;2. The use of NBT method and TBA were detected in patients with preeclampsia and normal pregnancy plasma SOD, MDA whether the expression level of statistical significance;3.Analysis of umbilical cord blood plasma AOPP and AOPP, MDA, SOD expression levels correlated AOPP clear whether the endothelial cells involved in inflammatory response in preeclampsia, from the molecular level of the mechanisms of preeclampsia.4. Analysis of 24-hour urine protein in patients with preeclampsia plasma AOPP level of correlation of plasma AOPP clear assessment of the role of disease severity;5.Statistics with SPSS 13.0 software, the data in that test of homogeneity of variance, the homogeneity of variance of data with One-Way ANOVA, LSD multiple comparison selection method; Heterogeneity of variance in the data using Welch test, multiple comparisons Methods Dunett T3 assay, P<0.05 when significant. Of AOPP and SOD, MDA, respectively, correlation analysis, correlation analysis of each group in the state analysis is done before testing, such as the parameters of normal distribution, pearson correlation analysis used for testing; if not normal distribution using the nonparametric Spearman correlation Analysis.Results1. Compared with normal pregnant women, patients with mild preeclampsia was significantly higher expression levels of AOPP, the difference was significant (P=0.003); compared with normal pregnant women, patients with severe preeclampsia were significantly higher AOPP, There was a significant difference (P=0.000), severe preeclampsia plasma AOPP level was significantly higher than mild patients (P=0.000);2. Compared with normal pregnant women, umbilical cord blood of patients with mild preeclampsia was significantly higher expression levels of AOPP, the difference was significant (P =0.001); compared with normal pregnant women, umbilical cord blood in patients with severe preeclampsia AOPP levels were significantly increased, the difference was significant (P=0.000), severe preeclampsia was significantly higher than plasma AOPP patients with mild (P=0.007)3.Compared with normal pregnant women, preeclampsia was significantly higher plasma MDA levels, the difference was significant (P<0.01); severe preeclampsia plasma MDA levels were significantly higher than that of mild patients (P<0.01); compared with normal pregnant women, preeclampsia plasma SOD level was significantly lower, the difference was significant (P <0.01); patients with severe preeclampsia SOD was significantly higher than mild patients (P <0.01.)4. By bivariate correlation analysis showed that patients with preeclampsia and 24 hour urine protein AOPP, there is a significant positive correlation (r=0.868, P=0.000); and cord blood AOPP were significantly positively correlated (r=0.638, P=0.000), and the expression of plasma MDA were significantly positively correlated (r=0.0.811, P=0.002), and SOD expression negatively correlated (r=-0.808, P=0.000).Conclusions1.Cord blood plasma and the expression levels of advanced oxidation protein products in patients with preeclampsia was significantly higher, and with disease severity is positively correlated, suggesting that increased maternal plasma levels of AOPP is an important pathological changes in preeclampsia might be involved in preeclampsia The occurrence and development.2.Preeclampsia expression of plasma AOPP and MDA were significantly positively related to negative correlation with SOD further clarify the occurrence of preeclampsia, the development of a variety of oxidation factors to participate and interact.3.Plasma AOPP levels and 24-hour urine protein was significantly positively correlated, suggesting that AOPP could be a new indicator for assessment of the disease.
