Objective:1 In clinical Shengmaiyin is often used to treat or aid in healing chronic heart failure. Shengmaiyin were recorded in 2005 version the 2010 edition of the People's Republic of China pharmacopoeia monographs first.. The extraction technology of Shengmaiyin's standard quality in pharmacopeia is ethanol extracting. While the traditional extraction process is boiled method. The author compares this two kinds of extraction process Shengmaiyin treatment of chronic heart failure rats extract effect. It provides theoretical basis of clinical application on Shengmaiyin.2 Use the low,middle,high doses of ethanol extraction in treatment of chronic heart failure rats, choose different levels of many experimental indexes of observation and testing, Shengmaiyin against the role of chronic heart failure mechanism and dose-respones relationship do preliminary studies. It Provides the theory basis for material foundation research and the secondary development on ShengMaiYin's treatment of chronic heart failure.Method:1 The method of medicine extract The prescription for extracting adopt the standard quality prescription of Shengmaiyin in pharmacopeia. The ethanol extraction method refers to the Shenmai Yin standard quality extraction way in pharmacopeia. While the boil is traditional method.2 Build the chronic heart failure model in rats.The male Wistar rats were abdominal injected ADR at a dose of 1.5mg·kg-1, twice a week for 7 weeks, total at a does of 21mg·kg-1 for model; the control group (n=6) received an equal volume of saline alone.3 Animal grouping and dosing method:The Wistar rats were randomly divided into six groups:the model group (n=10),masculine medicine group (n=8),low-does ethanol extraction group (n=8),mid-does ethanol extraction group (n=8),high-does ethanol extraction group(n=8),boil group (n=8). The control group without change. Since the 8th week, namely after stop injecting, giving drug 5 times at 8:00-9:00am once a day for 4 weeks. The masculine medicine group:giving captopril at a dose of 13.5mg·kg-1; low-does ethanol extraction group:giving at a does of 3mg·kg-1(as per medicinal materials unit); mid-does ethanol extraction group: giving at a does of 9mg·kg-1(as per medicinal materials unit); high-does ethanol extraction group:giving at a does of 27mg·kg-1(as per medicinal materials unit); boil group:giving at a does of 3mg·kg-1 (as per medicinal materials unit); control group giving equivalence distilled water; model group same as control group.4 The index and method for observation:4.1 During giving drug period, observe the appetite,hair color,posture,behavior signs and death detail of each rats group.4.2 After four weeks of giving drug, test each group rats hemodynamic parameters by biological function test systems.4.3 Before end the test, fasting on rats (ex water) for 12 hours. Then taking serum and viscera, weighing each organs and calculate each viscera coefficient.4.4 Taking the rats heart of each group, making pathological section to observe histological changes.4.5 Test the serum content on MDA,LDH,CD,SOD of each rats group according to the instruction of the MDA,LDH,CK and SOD.4.6 To adopt the ELISA to test the BNP in serum concentration of each rats group. The operation process refers to the instruction of BNP ELFA on rats ELIASA is applied to test the OD within 450nm and calculate the serum concentration of each rats group's BNP by standard curve.Results:1 The results comparison of different extraction technologyCompared with low-does ethanol extraction group, rats in boil group showed low spirit, movements reduced as well as food consumption The damage extent of muscular tissue in boil rats group is higher than low-does ethanol extraction group. Compare with model group, the low-does ethanol extraction group's serum BNP level reduced significantly so that near to control rats group level; the boil group's serum BNP level slight high than model group. This two kinds of extraction can reduce the liveness of MAD,LDH,CK in serum of chronic heart failure rats and improve SOD level. The SP,DP,AP,-dp/dtmax of model rats group improved significantly after compare with low-does ethanol extraction rats group and boil rats group.2 Different doses result comparisonThe general situation and behavior signs of low-does ethanol extraction group are same as masculine medicine rats group, and near to control rats group; the mid-does ethanol extraction rats group's status is better than model rats group; The general situation and behavior signs of high-does ethanol extraction rats group are familiar with model rats group. The death rate of high-does ethanol extraction rats group is high. Along with increase of does, the effect on chronic heart failure rats' hemodynamic parameters by ethanol extraction didn't show regular changes. That three experiments on ethanol extraction display:more does of drug, more damage on cardiac muscular tissue of rats. Along with increase of does, the effect on chronic heart failure rats' SOD and LDH by ethanol extraction didn't show regular changes, but the serum MDA,CK level showed regular changes.The serum BNP level improved as following the increase of does. The serum BNP level of high-does ethanol extraction rats group is higher than model rats group.Conclusion:1 The ethanol extracting technology is good than boil technology on healing chronic heart failure rats. But the percentage of extracting technology is higher than boil technology.2 The curative effect decreased along with increase the does of the ethanol extraction on healing chronic heart failure rats.
