Objective:We studied that TFPI-2 has differently express in gastric cancer tissues and normal gastric tissues,in order to investigate the mechanism of invasion and metastasis in gastric cancer. for judging the prognosis of gastric cancer and treatment of gastric cancer metasta- sis to provide scientific evidence.Materials and methods:64 specimens of gastric cancer postoperative patients were collected from the general surgery department of the affiliated hospital of Tai Shan medical university from November 2008 to June 2010. Specimens were taken for each cancer tissue and normal gastric mucosa (from the edge of cancer 5cm), a part of samples were kept in 10% formalin and then embedded in Paraffin. Other samples were collected and cut into pieces, snap frozen in liquid nitrogen, then kept frozen at -80℃.Expression of TFPI-2 protein were detected by Immunohistochemisty in gastric cancer.Expression of TFPI-2 mRNA were detected by RT-PCR.Result:Positive expression rate of TFPI-2 protein was 71.88% in normal gastric tissue,is significantly higher than gastric cancer 37.50%(P=0.000); Expression in no serosal invasi- on of TFPI-2 protein is higher than in serous invasion group( P=0.028). Expression in without lymph node metastasis higher than lymph node metastasis(P=0.008). TFPI-2 mRNA in gastric cancer tissue and normal gastric tissue were expressed , the difference was statistically significant(P=0.001) ; Expression in no serosal invasion of TFPI-2 mRNA is higher than in serous invasion group(P=0.000). Expression in without lymph node metastasis of TFPI-2 mRNA is higher than lymph node metastasis(P=0.000).Conclusion:Expression of TFPI-2 protein and TFPI-2 mRNA is lower in gastric cancer, Expression of TFPI-2 protein and TFPI-2 mRNA correlated with serosal invasion and lymph node metastasis.
