ObjectiveTo investigate the protective effect of 2,3,5,4′-tetrahydroxyl diphenylethylene-2-O- glucoside against damages of endothelial cells induced by lysophosphatidylcholine (LPC) and to study the underlying mechanisms.MethodsThe human umbilical vein endothelial cells(HUVEC) were treated with LPC (10ug/ml) for 24 h to establish the model of endothelial cells injury; HUVEC were prior exposed to 2,3,5,4′-tetrahydroxyl diphenylethylene-2-O-glucoside (10, 1.0 and0.1 umol/1) for 1 h, and then exposed to LPC in the presence of the drugs for 24 h in all. We also set the control group; At the end of the experiment, the cultured medium was collected for measuring the concentration of lactate dehydrogenase nitric oxide(NO), malefic dialdehyde(MDA) as well as asymmetric dimethylarginine(ADMA), and the cells were collected for measur- ing the intracellular reactive oxygen species(ROS) level and Cell survival and Apoptosis. The content of NO was measured by nitrate reductase method. The level of MDA was measured by thiobituric acid reaction(TBA) method. The concentration of ADMA was measured by ELISA. The intracellular ROS level was detected by Fluorospectrphotometer analysis. The intracellular Apoptosis rate was detected by flow cytometer. The intracellular Cell survival was detected by MTT.Results1. Compared with control group, exposure of endothelial cells to LPC (10ug/ml) for 24 h significantly increased the concentration of NO(P<0.05), MDA(P<0.05), ADMA(P<0.05) in the medium and the intracellular ROS level(P<0.05); concomitantly significantly decreased the concentration of NO(P<0.05) in the medium. 2. Compared with LPC group, 2,3,5,4′-tetrahydroxyl diphenylethylene-2-O-glucoside (10, 1.0 and0.1 umol/1) significantly inhibited the elevated concentration of NO(P<0.05) and MDA(P<0.05); ADMA(P<0.05) as well as the intracellular ROS level(P<0.05), concentration significantly attenuated the decreased level of NO(P<0.05) induced by LPC.Conclusionsl. 2,3,5,4′-tetrahydroxyl diphenylethylene-2-O-glucoside protected endothelial cells against damages by LPC.2. The protective effect of 2,3,5,4′-tetrahydroxyl diphenylethylene -2-o-glucoside on the endothelial cells is related to antagonise lipid peroxidation and to the level of ADMA.
