Objective:to study the anti-tumor Effect and Mechanism of Cardiac glycosides (digitoxin,digoxin, cedilanid) Methods:The human tumor Cells and normal cell tumor cells proliferation influence of Cardiac glycosides was test by MTT assay; With different concentration of the cardiac glycosides on HepG2 cell,Using hoechst33258 fluorochrome, the change of morphologic was observed by fluorescent microscope; Adopt Na+/K+-ATPase;with the different concertration of the cedilanid on HepG2 cell for different hours,the cell cycles were detected by flow cyrometry; Establish H22 mice subcutaneous tumor model and observe the effect of the cedilanid anti-tumor in vivo. Results:Cardiac glycosides can inhibit tumor cell proliferation, but to human normal liver cell HL-7702,it showed lower toxicity, through fluorescent Hoechst33258 testing,the results show that the cell morphologic changes HepG2 cells, 1h Abram role, after 6h, 12h typically seen in apoptosis morphological changes, as follows:part of cytoplasm narrowing, cytoplasm, nuclear chromosome highly concentrated aggregation and marginalized, nuclei in shape, the fractured pieces is typically seen crescent apoptotic body, and normal controls the nuclei of integrity. Conclusion:Cardiac glycosides can selectively affect the tumor cells proliferation, however, do not affect the human normal liver HL-7702 cell.Cedilanid induced G1 phase arrest of the cell cucle. Cedilanid had the effect of anti-tumor in vivo.
