Detection Of Brucella And Listeria Monocytogenes(LM) In Raw Milk By Duplex PCR ,antibody Screening On Four Major Animal Diseases In Hanzhong Area

Brucella is a Gram-negative ball-bacillus with a strongly pathogenicity to animals and cause a wide range of infections. Though no obvious clinical symptoms were presented in infected cow, but the bacteria still could be discharged by milk, and the food safety risk also was high to human by milk and dairy products. Listeria monocytogenes(LM) belongs to Gram-positive and is wide distribution in environment. LM is also a significant food poisoning micro-organisms and presents a high threat to human through food contamination. In recent years, the animal diseases presented increasingly forms in clinical, and it is difficult to prevent and control them. In order to control those diseases, the development trends should be predicted to assess the probability of disease outbreak. In view of this, according to the standardization of laboratory diagnostic methods, we detected the antibodies of highly pathogenic avian influenza, food-mouth disease, classical swine fever and Newcastle disease in Hanzhong area. The results showed:(1) Brucella and LM were detected in raw milk collected from 3 provinces(Yunnan, Gansu and Tianjin) in China by duplex PCR. 6 Brucella positive milk samples were found in787samples, the positive rate was 0.76%; no LM infected milk sample was found in 787 samples.(2) 695 sera, including to 246 pig sera, 130 bovine sera, 110 goat sera and 209 chicken sera were collected in Hanzhong region. The anti-FMD, CSF, AI and ND antibodies were detected from collected sera by ELISA. The results showed:For food-mouth disease O type: the 276 sera (77 pig sera, 114 bovine sera and 85 goat sera) were conformity to antibody level out of 360 sera (120 pig sera, 130 bovine sera and 110 goat sera copies were detected,which included 120 pigs,130 cattle, and 110 sheep), the antibodies pass rate were 64.2%, 87.73%, 77.3% respectively.Classical Swine Fever: 210 serum samples were conformity to antibody level out 246 sera ,the antibody passing rate was 85.4% .H5 subtype avian influenza: 206 sera were detected and 152 sera were antibody eligible, the pass rate was 73.8%.Newcastle disease: 209 chicken sera were detected and 155 were antibody eligible, the pass rate was 74.2%.The antibody pass rate of CSF, ND and AI meet the requirements of Department of Agriculture except the antibody pass of pig foot-mouth disease is slightly lower.
