Objective: To study the expression of OX40 and OX40L molecules in peripheral blood lymphocytes with systemic lupus erythematosus (SLE) patients and its possible mechanism of action in this disease. Regulating OX40L OX40 in target cells in corresponding expression and blocking the OX40 / OX40L pathways which can reduce inflammation happening can provide new ideas for the treatment of autoimmune disease.Methods: we analysed the expression of OX40 and OX40L level in lymphocytes by flow cytometric analysis detection techniques with 43 SLE patients and 15 cases of healthy volunteers. Meanwhile, we made comparison on the expression level of OX40 and OX40L in the peripheral blood lymphocytes between the nephritis type of SLE and without nephritis group . After SLE patients receiving a period of treatment ,we compared the expression level of the OX40 and OX40L in the peripheral blood lymphocytes in 10 cases patients.The specific steps of measuring the level of OX40 and OX40L: (1) 50ul blood were added to each flow tube by adding CD4-Tc, OX40-FITC and OX40L-PE, 4℃(time 20min); (2) setting negative control tubes: CD4-Tc, IgG-FITC and IgG-PE or CD19-FITC, IgG-PE were joined to controlling tubes, 4℃(reaction for 20min); (3) adding hemolysis agent cracking erythrocytes (4) catharsising with phosphat buffer solution (PBS) . (5) Centrifuging: 1800rpm, 5min. (6) After centrifugation,discarding supernatant, adding 0.5ml PBS ,we measured the phenotype by flow cytometry.Result: OX40 and OX40L in CD4 + T cells co-expressed. Compared with the control group: (1) SLE patients, CD4 + T OX40 + OX40L + cell subsets increased; (2) CD8 + OX40 + OX40L + cells subsets did not change; (3) OX40L + B cells were increased; (4) OX40L + monocytes increased; (5)The expression of OX40L in the NK cells was inhangced. In SLE patients, patients with nephritis type CD4 + OX40 + T cells increased significantly and much higher than non-nephritis-type; In addition, we found that the expression of OX40 and OX40L reduced in varying extent after treatment.Conclusion: SLE patients with T lymphocytes, monocytes, NK cells exist abnormal expression of OX40 and OX40L. With nephritis type ,the expression upregulated .There were great changes in the expression of OX40 and OX40L after treatment. Those suggest that OX40/OX40L and monocytes, NK cell interactions may contribute to the process of continuous activation of T cells, which participate in the occurrence of SLE development.
