Because of nice biological properties and excellent film formers of silk materials, it has extensive apply prospect in the therapy of the surface trauma which caused by fire, mechanic and chronic disease. While vascularization can't be fully achieved in tissue engineering is still the largest limit in the application of the three-dimensional scaffolds material. It's essential for the symplastic growth of the arteriole which is a main blood-supply artery in material while the capillaries have grown into the material step by step and developed the vessel network. The research on the regenerate process and mechanisms of arterioles had been considered as a new field for the past two decades. Further studies on arteriolar regeneration especially in biomaterial and tissue engineering fields should be paid more attentions. Based on such status, this paper aims to explore arterioles regenerate mechanism and the network construction with the capillaries in the silk protein biological materials. The arterioles growth and the process of the network construction between arteriole and capillary were investigated and researched in the situation of the biomaterial intervention in different tissue position.In this paper, the PSFF and the PVA sponges were implanted into the muscles of hind limb and back skin in rats, and the growth morphous and the quantity of arterioles in porous silk fibroin films (PSFF)material and the PVA sponges were observed and analyzed by tissue slices and SABC. In addition, the ultrastructure of arterioles and capillary, the location relations of the cells which composed the microvessels in PSFF were analyzed by TEM technology. The results showed that when the PSFF was implanted into the muscles of hind limb, the arteriole often appeared in the junction between material and tissue and then grown into the superficial zone of the material. The idea that the smooth cells of the regeneration arteriole were migrated from the terminal arteriole which connected with the arteriolization capillary was speculated. And the concrete development process and the mode were explored. By SABC and TEM observation, the SM cells of the arteriole in PSFF which implanted into the back skin in rats may originate from mesenchymal fibroblasts. The development process and concrete mechanism of the arteriole were analyzed in this situation by combing the paper reports and the experiments. Meanwhile some theoretical analysis was made about the network model between arteriole and capillaryThis paper is helpful for the further understanding of the vascularization of the material, and provided the theory basis and experiment foundation for the exploitation of the medical biomaterials which will be used more widely through studying on the regeneration mechanism of arterioles and network construction between arteriole and capillary in the silk fibro medical biomaterials.
