Objective To explore the value of the MRI application of RGD-Gd targeting fluorescent nano-probe in vitro LOVO cells and the possibility of RGD-Gd targeting fluorescent nano-probe against the cancer.Methods Cultured Colorectal cancer LOVO cells in vitro.Using nano-technology coupled the RGD peptide which was labled by quantum dot(abbreviated as QDs)and Gd3+,constructed RGD-Gd fluorescent nano-probe.The RGD-Gd fluorescent nano-probe,non-targeting of the QDs-Gd nanoparticles were incubated with LOVO cell 1h, at the same time ,establish no treatment control group.Then observe the binging situation through the Fluorescence phase contrast microscope ,MRI test T1 signal intensity,Flow cytometry analysis the cell cycle.Result The result of fluorescence phase contrast microscopecon,the RGD-Gd group can see fluorescence distributed in the surrounding the LOVO cell,the QDs-Gd group can see scattered fluorescence and the control group has no fluorescence distribution.The result of MRI showed the signal intensity of RGD-Gd group was higher than QDs-Gd group and control group(P<0.05).Flow cytometry showed RGD-Gd fluorescent nano-probe can block G2+S period of LOVO cell.conclusions: The RGD-Gd fluorescent nano-probe can specifically adsorbed on the surface of the colorectal cancer LOVO cells,can target enhanced the MRI imaging results,and may have some killing effect of tumor cells, provides some experimental evidence for anti-cancer therapy .
