Aim To examine the expressions of PDCD5 and Bcl-2 in cervical squamous cell carcinoma (CSCC) tissues. To study the antiprolifrative effects and the underling mechanisms of Glaucocalyxin A on human cervical carcinoma SiHa cells.Methods 89 CSCC tissue blocks and 25 normal cervical epithelium(NCE) tissue blocks were analyzed by immunohistochemistry for PDCD5 and Bcl-2 proteins.The antiprolifrative effect of Glaucocalyxin A on SiHa cells was measured by MTT assay. The morphological changes were observed by Hoechst33258 fluorochrome staining and transmission electron microscope (TEM). The rates of cell cycle distribution and apoptosis were analyzed by flow cytometry. The molecular mechanisms involved in the effects of Glaucocalyxin A on SiHa cells were studied by RT-PCR and Western blot.Results There were significant differences of PDCD5 expression between CSCC tissues (33.71%,30/89) and NCE tissues (76%,19/25).The expression rate was associated with pathological grade(P﹤0.05) and lymph node metastasis(P﹤0.001). There were also significant differences of Bcl-2 expression between CSCC tissues (67.42%,60/89) and NCE tissues (40%,10/25).The expression rate was associated with pathological grade(P﹤0.01) and infiltration depth(P﹤0.001). The expressions of PDCD5 protein were negatively related to those of Bcl-2 protein in CSCC tissues(r=?0.427, P﹤0.01).The growth potential of human cervical carcinoma SiHa cells was siginificantly inhibited by Glaucocalyxin A. Apoptosis morphological changes were observed on SiHa cells after Glaucocalyxin A treatment by Hoechst 33258 fluorochrome staining and TEM."Sub-G1"phase peak was appeared and cell cycle was arrested in S phase after Glaucocalyxin A treatment. The expression levels of PDCD5,p53,Bax,Caspase-3 and Caspase-9 mRNA were upregulated and the expression level of bcl-2 mRNA was downregulated. The expression levels of PDCD5 and Bax proteinswere upregulated and the expression level of Bcl-2 protein was downregulated. Conclusions PDCD5 and Bcl-2 can serve as a biomarker to predict the malignant degree of CSCC.The antiproliferative effects of Glaucocalyxin A on human cervical carcinoma SiHa cells may be associated with apoptosis and S phase cell cycle arrest through Mitochondrial apoptosis pathway.
