Objective To observe the expression levels of AMP-activated protein kinaseα2 (AMPKα2) and peroxisome proliferator-activated receptorγ(PPARγ) in adipose tissue of type 2 diabetes with metformin treatment, and the influence on glucose and lipid metabolism to discusses the mechanism to improve insulin sensitivity.Methods After 30 male wistar rats feed adaptability one week with ordinary feed, randomly divided into normal group (NC, n = 10) and diabetic group (DM, n = 20). NC group were given ordinary feed to the end of the experiment. DM group have given high fat feed for 4 weeks later, high- fat diets combined by left celiac injection of STZ 40mg/kg one-time in small doses to prepare type 2 diabetes model. Five days later, measured the tail blood with blood glucose meter, select FPG≥16.7 mmol/L as successful mold. The diabetic rats model were randomly divided into diabetic group (DM-C,n=9) and treatment group (DM-T,n=9). The DM-C group and DM -T group were continued to give high-fat feed, meanwhile DM-T group were given hydrochloride metformin orally once daily for 4 weeks. NC group and DM-C group were given volumetric 0.9% physiological saline to irrigate stomach for 4 weeks. The final statistics : 2 of 30 rats were removed because their glucose levels don't mach the standar of model rats. 2 of 30 rats were dead during the process of irrigating stomac, 26 rats were survived to the end of the experiment , they are 10 rats of normal group, 8 rats of diabetic group, 8 rats of treatment group. Weight of rats was measured before and after treatment, at the end of the experiment the weight of the rats'adipose in perirenal and testis were measured. Then to calculate of total body weight ratio of fat . Their serum were used to detect of Fasting plasma glucose (FPG) , Fasting insulin (FINS), and Fasting sensitive index ( INI) were calculated. serum total cholesterol ( TC ) , triglycerides ( TG ) , high density lipoprotein(HDL-C),Low density lipoprotein(LDL-C) levels also were detect; real time - polymerase chain reaction (Real Time -PCR) to detect the expression of AMPKα2 and PPARγmRNA in adipose tissue.Results 1 weight after Feeding by high fat diet four weeks and small dose STZ intraperitoneal injection , before the treatment with metformin, the weight of the rats in DM group increased significantly than NC group rats(P<0.05), after the Treatment for 4 weeks the weight of the rats between the DM-C group and the DM-T group ,the decline of the weight of DM - T rats were reduced significantly(P<0.05).2 weight of adipose tissue in perirenal and testis and total body weight ratio of fat compared with NC group, the weight of the adipose tissue in perirenal and testis and the total body weight ratio of fat of the DM - C Group were significantly decreased (P < 0.05) compared with DM-C group, the weight of the adipose tissue in perirenal and testis and the total body weight ratio of fat of the DM-T Group were significantly increased (P < 0.05) .3 Fasting plasma glucose (FPG) , Fasting insulin (FINS), and Fasting sensitive index ( INI) compared with NC group, Fasting plasma glucose of DM-C Group and DM-T Group were significantly increased (P < 0.05)Fasting insulin (FINS), and Fasting sensitive index ( INI) of DM-C Group and DM-T Group were significantly decreased(P < 0.05);compared with DM-C group, Fasting plasma glucose of the DM-T Group were significantly decreased(P < 0.05), Fasting insulin (FINS), and Fasting sensitive index ( INI) of the DM-T Group were significantly increased(P < 0.05).4 blood lipid compared with NC group, serum total cholesterol(TC), triglycerides(TG), Low density lipoprotein(LDL) levels of DM-C Group were significantly increased(P < 0.05),high density lipoprotein(HDL) of DM-C Group were significantly decreased (P < 0.05);compared with DM-C group, serum total cholesterol(TC), triglycerides(TG), Low density lipoprotein(LDL) levels of DM-T Group were significantly decreased(P < 0.05),high density lipoprotein(HDL) of DM-T Group were significantly increased(P < 0.05).5 expression of AMPKα2 and PPARγmRNA in adipose tissue compared with DM-C group, the expression of AMPKα2 and PPARγmRNA in adipose tissue of DM-T Group were significantly increased(P < 0.05).Conclusion: 1 Metformin can increase the expression of AMPKα2 and PPARγmRNA in adipose tissue of Type 2 diabetic rats.2 Metformin can improve the abnormalities of the lipid metabolic, increases insulin sensitivity of type 2 diabetic rats.3 Metformin can inhibit the decompose of adipose tissue in perirenal and testis.
