Pulmonary hypertension is a resting mean of pulmonary artery average pressure> 3.33kPa (25mmHg) or movement>4kPa (30mmHg).The two basic pathological changes of pulmonary artery hypertention (PAH) are pulmonary artery(PA) contraction and pulmonary artery vascular structural remodel. Inhibiting the pulmonary vasoconstriction, deteriorating the structural remodeling of pulmonary vascular, improving pulmonary hemodynamics and preventing pulmonary vascular smooth muscle cell from proliferation are the key of treatment.Apoptosis is a kind of biology action of cells, it plays an essential role in abnormal cells. It plays important role in the evolution of biology, the stability of internal environment and the development of multiple systems. Apoptosis is not only a kind of special type of apoptosis, and it also has important biological significance and complex molecular mechanisms.Taurine (Tau) sources from food, almost resides in all animals' cells by means of liberation, it's an endogenous anti-damage material in organism, it have cytoprotective action such as to accommodate cells' calcium homeostasis, stabilize the cell envelope and to clear the free radical et.al. It also has effect on induces apoptosis in some kinds of cells.Objective:To study the effect of Taurine on apoptosis in PASMCs, and whether the mitochondrial-dependent pathway and death-receptor pathway act in the mechanism.Methods:Culture the PASMCs, and divided the cells into control, SD, and Taurine (Taurine high,medium and low dose). The cells were subject to growth arrest for 24h before placed into 20%FBS-containing DMEM or treated with serum deprivation medium or treated with different concentration of Taurine for next 24h. MTT assay, Acridine orange(AO) assay, Mitochondrial depolarization assay and western-blot analysis on the expression of Bax,Bcl-2,Procaspase-9,Procaspase-3 and Fas were used to study the mechanism.Result:A major finding of this study is that the Taurine effects many apoptosis index, such as decreasing the cell viability in a concentration manner, decreasing the expression of Procaspase-3, Procaspase-9,and Bcl-2, increasing the expression of Bax, accrescencing the mitochondrial depolarization, causing the nuclear shrinkage, all these datas demonstrated that Taurine induced the apoptosis in pulmonary artery smooth muscle cells through mitochondrial-dependent pathway.Conclusion:our results have showed that Taurine induces apoptosis in pulmonary artery smooth muscle cells through mitochondrial-dependent pathway and death-receptor.
