Objective : Construction of a new splice variants of CD44 siRNA expression plasmid to obtain plasmid was stably transfected in human nasopharyngeal carcinoma 5-8F cell lines for the study of CD44 splice variants in nasopharyngeal carcinoma of new features in the experimental basis.Methods : The synthetic oligonucleotide strand annealing to form double-stranded, ligated into pGenesil-1.3-kan/Neo expression vector and sequencing. Liposome, the recombinant plasmid to the 5-8F cells by G418 selection, filter paper method to obtain monoclonal cells, the establishment of stable transfected 5-8F cell lines.Results: Construction of a new splice variants of CD44 siRNA eukaryotic expression vector, obtained after G418 selection of monoclonal cell lines transfected 5-8.Conclusions: Successfully constructed a new splice variant CD44 siRNA eukaryotic expression vector and successfully transfected into 5-8F cells and obtain a monoclonal cell line.
