Objective:To investigate the expression of tumor infiltrating lymphocytes (TIL) in primary colorectal carcinoma and prognostic significance.Methods:Immunohistochemical technique (IHCA) and flow cytometry (FCM) were used to detect the expression of tumor-infiltrating lymphocyte subpopulation (CD3,CD4,CD8,CD45RO,CD25,CD28) and HLA-DR antigen in 60 cases of primary colorectal carcinoma.The relationship of their expression with clinico-pathological feature and prognosis of colorectal carcinoma were analyzed.Results:The expression rats of tumor-infiltrating lymphocyte subpopulation (CD3, CD4, CD8, CD25, CD28 and CD45RO) and HLA-DR antigen were respectively 63.3% (38/60),28.3%(17/60),41.6%(25/60),25.0%(15/60),36.7%(22/60),38.3%(23/60) and 41.7%(25/60) by immunohistochemical staining. The expression of CD4, CD8, CD28 and HLA-DR antigen were no correlations with patients'gender,age,lymph node metastasis and differentiation degree of tumor and were correlations with tumor Dukes stage and prognosis of patients.Their expression rats were decreased with Dukes staging increasing and were significantly higher in≥5-years survival group than≤3-years denth group, there were significant differences. (P<0.05).The expression rats of CD3 and CD25 were correlations with tumor Dukes stage,lymph node metastasis and prognosis of patients and were no correlations with patients'gender,age and differentiation degree of tumor.There was no correlations with clinico-pathological feature and prognosis of colorectal carcinoma for the expression of CD45RO.Flow cytometry detected the expression of CD3, CD4, CD8, CD25, CD28 and HLA-DR antigen. The expression rats of them were significantly higher in≥5-years survival group than≤3-years denth group, and there were significant differences. (P<0.05). The expression of them were correlations with prognosis of patients.Conclusion:TIL subpopulation(CD3, CD4, CD8, CD25 and CD28) are the important characteristic markers in colorectal carcinoma, which relates With the prognosis, and they are of clinical significance to raise anti-tumor immune responses.
