Objective:To observe the effect of zinc of different concentrations on the develop-ment of murine embryonic palatal shelve,and chose a proper concentration of zinc which can promote the normal development of embryonic palatal shelve;To establish all-trans retinoic acid-induced mice model with cleft palate and explore the antago-nistic action of zinc on the all-trans retinoic acid-induced cleft palate model.Method:The embryonic palatal shelves were isolated and cultured in vitro at E13. The embryonic palatal shelves were divided into A,B,C,D,E,F group accordin g to different interventional factors,embryonic palatal shelves of A,B,C,D,E,F group were treated with 0μmol/L zinc,50μmol/L zinc,75μmol/L zinc,95μmol/L zinc, 10μmol/L all-trans retinoic acid and 10μmol/L all-trans retinoic acid+75μmol/L zinc respectively.The embryonic palatal shelves was collected after 24h and 48h, surface structure of mouse embryonic palate was observed with Stereo microscope and the embryonic palate fusion rate was recorded,moreover,the process of embryonic palate fusion after 48h was observed by HE staining.Result:The fusion rate for A,B,C,D,E,F group were 27.8%,53.9%,85%,3 2%,0% and 6.7%respectively.There were no statistical significance among A,B,D group by Chi square test(P>0.05),but statistical significance was declared between C and other groups(P<0.05).The C group was chose as the proper concentration for its eff-ect on embryonic palate fusion.Compared E with F group,although the rate of cleft palate in E group was lower than that of F group,no statistical significance was de-tected between them(P>0.05).Conclusion:Embryonic palatal cultured in vitro can be repeated the palatal fusion process in vivo and effect of Zinc on the growth of Embryonic palatal was observ-ed.Zinc of 75μmol/L can promote the fusion of Embryonic palatal,which was chose as the proper concentration of zinc.All-trans retinoic acid-induced cleft palate model was established successfully.No Embryonic palatal fusion was observed when treated with all-trans retinoic acid (10μmol/L) and a few Embryonic palatal fusion can be detected when treated in combination with proper zinc concentration, but there was no statistical significance between them.
