Objective:To explore ethylhexyl phthalate (DEHP) of endocrine factors and its' metabolites (single-ethylhexyl phthalate, MEHP) whether there is injury to spermatogonia in young male Wistar rats.Methods:The 96 young male Wistar rats in 2 week after birth were randomly divided into 8 groups (12 each group). To lavage normal saline (0.9% NS 0.2ml·kg-1·d-1, feeding 3 weeks) in young male Wistar rats as a negative control group (NC) and cyclophosphamide (CTX 100mg·kg-1·d-1, feeding 1 week) as a postive control group(PC). To lavage respectively DEHP and MEHP at low dose (100mg·kg-1·d-1, feeding 3 weeks), middle dose group (200mg·kg-1·d-1, feeding 2 weeks), high dose group (300mg·kg-1·L d-1, feeding 1 week)in the rest groups.To examine testicular tissue changes in different times and in different doses. Whats' more, to examine spermatogonia apoptosis in testicular tissue by TUNEL,the expression of c-kit and PI3K by immunohistochemistry, sex hormones level(follicle stimulating hormone FSH, luteinizing hormone LH and testosterone T) in serum by radion immunoassays.Results:(1),The atrophy,development retardation and amount decreasing of spermatogonia in medication groups and vacuoles and engorge of its'mitochondria whereas there is no significant difference between spermatogonia injury of high dose medication group in short time and low dose in long time. (2),Spennatogonia apoptosis was increasing in medication groups (P<0.01),also between high dose medication group in short time and low dose in long time(P> 0.05).(3),To compared with the control group, the expression of c-kit and PI3K was decreasing in medication groups (P<0.01), also between high dose medication group in short time and low dose in long time(P> 0.05).(4),The FSH and LH levels in serum sex hormone was increasing in medication groups whereas T was decreasing (P<0.05), also between high dose medication group in short time and low dose in long time(P> 0.05).Conclusion:The DEHP and its metabolites (MEHP) may markedly induce injury in spermatogonia in young male rats and may damage spermatogenesis in children. And children should avoid excessive exposure to DEHP and its metabolites MEHP.
