Objective:To elucidate the relationship of the expression of chemokine receptors CXCR4, CXCR5, CCR7 and their ligands, chemokine CXCL12, CXCL13, CCL21, in 5 cases of primary central nervous system lymphomas and their clinical significance.Method:The expression of chemokine receptors in 5 cases of PCNSL and 10 normal brain tissues were investigated using immunohistochemical and reverse transcription polymerase chain reaction (RT-PCR) methods.Results:1. Immunohistochemistry:Of the 5 PCNSLs, all specimens (100%) were positive for CXCR4, CXCR5, CCR7, CXCL12, CXCL13 and CCL21. Of the 10 normal brain tissues, all specimens (100%) were positive for CXCL12, and none was positive for CXCR4, CXCR5. CCR7, CXCL13 or CCL21.2. RT-PCR:Of the 5 PCNSLs, all specimens expressed the RNA of CXCR4. CXCR5, CCR7, CXCL12, CXCL13 and CCL21. The specimens of normal brain tissue were only expressed the RNA of CXCL12.Conclusion:Chemokine receptor CXCR4, CXCR5, CCR7 bind to their respective ligands, chemokine CXCL12, CXCL13, CCL21, play important roles in the pathogenesis of primary central nervous system lymphoma.
