Background and objective:The extractive of Hypericum Perforatum L (TEHP) is a kind of Chinese medicine prepared in our laboratory. The main active ingredient of TEHP is hypericin extracted from Hypericum perforatum L. Preliminary research of hypericin anti-C VB3 induced viral myocarditis had been performed by our subject group. In this paper the anti-autoimmune myocarditis effect and mechanism of TEHP were studied by establishing cardiac myosin induced myocarditis model, which provided animal experimental data for the future clinical application.Methods:1. Use supercritical CO2 fluid extraction technology to extract hypericin (the main active ingredient of TEHP) and determine its content by HPLC. Response surfacedesign (RSD) was applied to optimize the conditions for the extraction of hypericin, including extract ive time, extractive pressure and entrainer concentration.2. To Perform acute toxicity test in confirming the toxic action for oral administration. 3. To establish autoimmune myocarditis model in study the effect and mechanism of TEHP.Results:1. The optimized condition of supercritical CO2 fluid extraction was at 50.3℃,25.29 MPa,2 L/min for 2 hours with supercritical CO2,0.5 mL/min ethanol as dynamic cosolvent and 78.6% ethanol-water solution as entrainer. The content of hypericin reached 1.087%.2. The oral maximum dosage of TEHP was 24 g/kg, the toxic action of it is very low.3. TEHP could decrease the expression of PFP/Gram B in myocardium, reduce the amount of NO in mouse myocardial tissue and the amount of MDA in mouse serum, and increase the vitality level of SOD in body. Meanwhile decrease CD4/CD8 of T lymphocytes in peripheral blood. The possible anti-autoimmune myocarditis mechanism of TEHP was related to PFP/Gram B pathway and could protect the body from free radicals damage by improving antioxidant capacity.Conclusion:TEHP played a good therapeutic effect on autoimmune myocarditis. It could not only reduce the inflammation in autoimmune myocarditis, but also relieve immune injury on myocardium. TEHP was of great potential to be developed as the therapeutic drug for autoimmune myocarditis.
