| Objective To study the effect of different theraputic methods on airway inflamma-tion and remodeling in the mice asthmatic model.Methods 60 healthy female Kunming mice were divided into six groups randomly, 10 mice of each group, normal control group (A),asthma group (B),Pulmicort group (C),Ginseng-Schisandrae Drug decoction group(D),Xiao qinglong decoction group (E),two step sequential method group(F).The mice of B,C,D,E,F groups were established the models of asthma using ovalbumin,after building,each group was given different therapy.Then observe the pathological change in lung tissue.Finally assay the levels of IL-4,INF-γin BLAF with enzyme-linked immunosorbent assay (ELISA) and detect the level of MMP-9 and TIMP-1 of lung tissue with immunohis-tochemical analysis.Results①There were no wheezing symptoms in group A while there were wheezing symptoms in other groups and the wheezing symptom of group B was more serious than the treatment groups.There was no significant difference among the treatment groups.②Pathological analysis of the lung showed that group A had a little inflammatory cell infiltration,which was significantly lower than that of other groups(P<0.05) and group B had a large number of inflammatory cells infiltrating around tracheas, which was significantly higher than other groups(P<0.05); There was no significant difference among the treatment groups(P>0.05).③there was no significant difference between group A and group F(P>0.05); there was significant difference among other groups (P<0.05);④The level of IL-4 in BALF was significantly lower in group A than that of other groups (P<0.05) while the level of IL-4 in BALF was significantly higher in group B than that of other groups(P<0.05) and there was no significant difference among the treatment groups(P>0.05).⑤The level of TIMP-1 of lung tissue was lower in group A than that of group B,D (P<0.05) and group B was higher than that of other groups(P<0.05), group D was higher than group E(P<0.05), which had significant difference among the above groups (P<0.05); there was no significant difference among other groups(P>0.05).⑥The level of MMP-9 of lung tissue and the ratio of MMP-9 and TIMP-1 were lower in group A than that of other groups while that of group B were higher than other groups (P<0.05); there was no significant difference among the treatment groups.Conclusions①The model of asmatic mouse stimulated by OVA may be resultd from the unbalance of Th1/Th2 by detecting the unbalance of IL-4 and INF-γwhile the unbalance of MMP-9/TIMP-1 may demonstrate that there was airway remolding at early period.②Traditional chinese medicine may reduce airway inflammation and airway remolding by adjusting the expression of the unbalance of Th1/Th2 and MMP-9/TIMP-1 respectively, which can be got the same result by using pulmicort. |
