To investigate the hepatoprotective effect of silymarin on cyclophosphamide induced liver injury and its possible mechanism.Hepatocytes of suckling mice were isolated and cultured. Cell viability, intracellular enzymes leakage, GSH and MDA contents of hepatocytes were measured. The serum biochemical parameters , oxidative parameters and anti-oxidative parameters of liver tissue of mice inoculated with S180 cells were measured. The pathology of the liver was observed with microscope. The weight of S180 sarcom was weighted.The elevation of MDA content of hepatocytes and LDH,ALT level in supernatant of hepatocytes induced by CP were significantly reduced by silymarin. The elevation of serum parameters, MDA content of liver tissue,the less of GSH content and CAT,SOD activities induced by CP were restored remarkably by silymarin. Spotty and focal necroses of hepatocytes in the liver of CP group were found with a great deal of inflammatory cells infiltration, while the damage in the liver of silymarin group was ameliorated obviously; The combination of high-dose silymarin with CP were found to be more potent than CP alone in antineoplastic effect. Silymarin could alleviate CP-induced liver injury. The hepatoprotective mechanism of silymarin maybe due to anti-oxidative activity.
