| Objective To identify gene expression pattern of steroid-induced femoral necrosis(SINFH) in rats by gene chip, try to find out the initiating factors and molecular mechanisms.And then to treat with Youguiyin, to observe the regulation of Youguiyin on the gene expression pattern of SINFH in rats,and to investigate the molecular mechanism of Youguiyin prevention and treatment of SINFH.Methods All the 30 rats were randomly divided into control ,experimental groups and youguiyin group (n=10 in each group). The rats were injected intraperitoneally with endotoxin,and then injected intramuscularly with high-dose methylprednisolone or saline in experimental group and youguiyin group.The control group were injected with saline.The rats received a daily intragastric administration of youguiyin in youguiyin group,and 0.9% Nacl in the model and normal groups.6 weeks later, mRNA was extracted from the femoral head of rats in the every groups,and the cDNA probes were obtained by inverse transcript,then carried out microarray detection. The quantitative RT-PCR was used to confirm the results of the mieroarray,and the differentially expressed genes were also analyzed for the functional annotation by GO(gene ontology).Results Compared to the control group,190 genes in the experimental group were differentially expressed, with 52 up-regulated and 138 down-regulated. Of these genes,102 are known (have deposited in GeneBank), while 88 of them are unknown. The known genes can be divided into several families according to their biological functions, such as:oxidative stress, apoptosis, signal transduction, angiogenesis, extracellular matrix, lipid metabolism, and gene transcription related genes. The results of quantitative RT-PCR are consistent with gene-chip results. Compared to the model group,92 genes in the youguiyin group were differentially expressed.These genes were mainly assigned to signal transduction,metabolic,transport,apoptosis,cell proliferation,blood coagulation,cell cycle and antioxidant activity.Conclusion Our findings indicate that many genes which are involved in different signaling pathways were differentially expressed between SINFH rats and normal rats. Our findings suggest that the development of SINFH is a complicated and dynamic process affected by multiple factors and signaling pathway, and regulated by various genes. Youguiyin can control gene expression of SINFH and cell signaling processes inside and outside of cell. This may be the molecular mechanism of Youguiyin prevention and treatment of SINFH.These investigations explained the efficacy mechanism of traditional Chinese medicine prevention and treatment of SINF from the perspective of gene expression and enrich theories of kidney governing bones "... |
