ObjectiveTo observe the effect of Intermedin (IMD) on the pancreatic tissue of acute pancreatitis(AP)rats, and then to explore the protective effect of IMD on the pancreatitis in the rat, by intraperitoneal injecting IMD in the rat model of acute pancreatitis.Methods①Selected healthy male SD rats, weighing250g-300g, then randomly divided into four groups:normal control (NS) group, acute pancreatitis (AP) group, adrenomedullin (ADM) treatment group, intermedin (IMD) treatment group, eight in each group, Prepared AP models by subcutaneously injection of caerulein (100μg/kg),2hours after modeling, intraperitoneal injection of ADM, IMD (7nmol/kg) was given to the two treatment groups, then killed the rats6hours after modeling;②Compared the changes of pancreatic tissue in each group by the way of pancreatic histopathological score;③etected the degree of edema in pancreatic tissue by drying method;④etected the changes of malondialdehyde (MDA), amylase (AMY), nitric oxide (NO), inducible nitric oxide synthase (iNOS), endothelial nitric oxide synthase (eNOS), superoxide dismutase (SOD)in the serum;⑤Detected the changes of inducible nitric oxidesynthase (iNOS) and endothelial nitric oxide synthase (eNOS) in the pancreatic tissue by the application of Western blotting.Results①The results of pathology score:AP group>ADM group>IMD group>control group, and the differences between each group were statistically significant (P<0.01);②The degree of edema in the pancreatic tissues:AP group>ADM group>IMD group>control group, and the differences between AP group and the two treatment groups with the control group were significant (P<0.05), there was no difference between the two treatment groups (P>0.05);③The results of MDA, AMY, NO and iNOS in serum:AP group>ADM group> IMD group>control group, and the differences between each group were significant (P<0.05); the results of eNOS in serum:AP group<ADM group<IMD group<control group, and the differences between each group were statistically significant (P<0.01); the results of SOD in serum:AP group ADM group<IMD group<control group, and the differences between each group were significant (P<0.05);④The results of Western blot: the content of iNOS protein, AP group>ADM group>IMD group> control group, and the difference between each group was statistically significant (P<0.01); the content of eNOS protein, AP group<ADM group<IMD group<control group, and the difference between each group was statistically significant (P<0.01).ConclusionIMD can lower MDA and increase SOD to alleviate the pancreatic oxidative stress injury in pancreatitis, in the same time, It can increase the pancreatic exocrine function and reduce the production of inflammatory mediators by regulating the expression of iNOS and eNOS, which making the protective effects on pancreatic tissue, the improvement of pathology and the extent of edema in pancreatic tissue, and the protective effect on the pancreatic tissue is more obvious than ADM.
