[Objective]The aim of the study is, based on the effective applying strengthening qi, nourishing yin,quyu to treat SS, through studying its impact on Spontaneous models, spleen index, submandibular Glande Index and blood serum IFN-γ and IL-10,to explore the possibility of using strengthening qi, nourishing yin,quyu to cure SS.[Methods]1. We selected60NOD mice and10normal are chosen, all of which are eight weeks old, female, SPF level, and are raised under the condition of constant temperature conditions without special bacteria (Shang slac laboratory animal CO.LTD).NOD mice and10normal ICR mice were randomly divided as six groups:control group, TCM low does group,TCM middle does group,TCM high does group, HCQ group and Chinese-western medicine group. ICR group consists ten normal mice, which is the reference group. During the experiment, the amount of drinking water of mice is measured. Before feeding the water, each groups bottles' weight is measured. After24hours, the total amount of each groups' drinking bottle is measured. The difference of the two amounts is the amount of the drinking water amount of the mice each day. At the end of the eighth week, the blood from all NOD mice's arteriae femoralis are fetched. The serum IFN-γ IL-10were measured by ELISA method in60NOD mices and10normal mices (normal control group). mice are killed after its femoral artery's blood is fetched, spleen and submandibular Glande is fetched and weighed,and the index of spleen and index of submandibular Glande is counted.a part of submandibular Glande for detecting the mean optical density by immunhistochemistry,and the other part be used to abserve its pathological changes under light microscopic.[Results]3.1The test results of Serum of IFN-y and IL-10:Compared to normal control group, the expression level of IFN-y and IL-10was significantly higher in model group (P<0.01); Low-dose group and model group has no significant difference (p>0.05); The expression level of IFN-y and IL-10in the rest of the treatment group was significantly decreased than the expression in model group (P<0.01).3.2The test results of Submandibular gland and spleen index:Compared to normal control group, the index of Submandibular gland and spleen was significantly increased in model group (P<0.01); Low-dose group and model group has significant difference (p<0.05); the index of Submandibular gland and spleen in the rest of the treatment group was significantly decreased than the expression in model group (P<0.01).3.3The results of amount of drinking water:The amount of drinking water of the model group were significantly higher than the normal group and treatment group(P<0.01); the amount of drinking water of model group were significantly higher than the normal group; Compared to model group, water intake in treatment group was significantly decreased(P<0.01).3.4Average optical density of positive particles used by immunohistochemical: Compared to normal control group, the expression level of IFN-y and IL-10was significantly higher in model group (P<0.05); the hydroxychloroquine group and model group has no significant difference (p>0.05); Compared to model group, treatment group was significantly decreased(P<0.05); Integrative Medicine group and high-dose group was significantly decreased (p<0.01).4. Conclusion(1) Compared with the normal control groups, IFN-gamma, IL-10cytokine levels in the submandibular gland and the serum was significantly increased in the model group, and the immune system was abnormally activated.(2) Strengthening qi, nourishing yin,quyu can significantly reduce the serum level of NOD and the IFN-gamma, IL-10.(3) Strengthening qi, nourishing yin,quyu regulated the balance of Thl/Th2cells by reducing IFN-gamma, IL-10expression in serum and in the submandibular gland to reduce the damage of the submandibular gland secretory function.
