Background and Aims:Visceral high sensitivity plays an important role in the pathogenesis of the irritable bowel syndrome (IBS), probiotics can regulation visceral sensitivity and improve IBS symptoms. The aims were to investigate the effect of microecological preparation Peifeikang in visceral sensitivity of IBS rat models, and to elucidate the possible mechanism of Peifeikang in the treatment of IBS.Methods:A total of28SD rats were randomly divided into four groups:normal control group, IBS group, DC group and Peifeikang-treated group. IBS model was induced by acute bound stress method. Mast cells degranulation of colonic mucosal tissue was observed with improved toluidine blue staining. The levels of colonic tissue and serum histamine,5-HT were measured by enzyme-linked immunosorbent assay(ELISA). The visceral sensitivity was assessed according to the abdominal wall evacuated reflection (AWR) score. The TLR2gene expression level in rat colonic mucosa was detected by RT-PCR. Results:Compared with the normal control group, the number of mast cells and degranulation ratio of colonic mucosal and the levels of colonic tissue and serum histamine,5-HT visceral sensitivity and the TLR2gene expression level were significantly higher (P<0.01). The above indexes in Peifeikang-treated group were significantly lower than those in IBS group (P<0.01). There were no significant difference among Peifeikang-treated group, normal control group and DC group (P>0.05)Conclusions:The microecological preparation Peifeikang can decrease visceral high sensitivity in IBS rat models. The mechanism may be related to inhibiting colonic mucosal mast cells degranulation, reducing histamine,5-HT level in colonic tissue and blood and degraduating TLR2gene expression level.
