Objective:To study the allele frequency distribution and the genotypic frequency of the rs7903146 and rs290487 of TCF7L2 gene,and to investigate the association of transcription factor 7-like 2(TCF7L2)polymorphisms with the early phase function ofβ-cell in type 2 diabetes mellitus(T2DM).Methods:The rs7903146 and rs290487 of TCF7L2 was genotyped in 213 Hans around Qingdao district (99 patients with type 2 diabetes mellitus and 114 normal subjects) by PCR—restriclion fragment lengh polymorphism(PCR—RFLP).The early phase function ofp-cell of the 99 patients were detected in L-arginine (L-ARG)stimulation test.Results:(1)In T2DM group, C/T genotype frequecies and T allele frequency of rs7903146 were signifcanly higher than those in control group (χ2=4.678 P=0.031;χ2=4.507 P=0.034). The C/T genotype and the T allele frequency of individuals with a family history in T2DM group were signifcanly higher than those in control group (χ2=4.896 P=0.027;χ2= 4.739 P=0.029), and had no significant deviation with the individuals without a family history (P>0.05). The C/T genotype can significantly increased the risk of of T2DM than the CC genotype (OR=3.28,95%CI为1.026—10.459,P=0.031)。(2) In T2DM group, the early phase function ofβ-cell of C/T genotype and C/C genotype of rs7903146 had no significiant disparity (3) CC,CTandTT genotype frequecies and T allele frequency of rs290487 had no signifcanly deviation with those in control group (χ2=0.037, P=0.847>0.05;χ2=0.037, P=0.847>0.05). The genotypes and the T allele frequency of individuals with a family history in T2DM group had no signifcanly deviation with those in control group (x2=0.162 P=0.922>0.05;χ2=0.156 P=0.693>0.05).Conclution:The rs7903146 polymorphism of TCF7L2 is a risk factor of the type 2 diabetes in Qingdao district,but not with the early phase function of the P-cell; The rs290487 polymorphism of TCF7L2 is not a risk factor of the type 2 diabetes in Qingdao district.
