Objective:To observe the differences of the number of hippocampal neurons,hippocampal volume,cell volume during kindling group in rats with temporal lobe epilepsy,non-kindling group and control group in rats, to explore the causal relationship of hippocampal sclerosis with Temporal lobe epilepsy.Methods:Status epilepticus was induced by a 20 min stimulation of the amygdala, Stimulated rats were monitored with a video recording system for the 3-months follow-up period to record the appearance of SRS to establish the post-status epilepticus model of temporal lobe epilepsy in rat. to use optical disector estimate the number of pyramidal cell, Cavalieri methods for volume of pyramidal layers, Nucleator for average volume of pyramidal cell. Statistically to analysis the difference of pathological changes for pyramidal cell during three groups rats.Results:the average neuronal number of rats during control group,non-kindling group and kindling group respectively were:616.4±212.46×103,635.0±249.53×103,875.3±149.64×103,there is no statistical difference between the groups (P>0.05), hippocampal average volume: 58267±20927.13×105μm3,56773.8±13983.93×105μm3 60722.8±8154.09×105μm3, no statistical difference between the groups (P>0.05). Neuronal average volume:2515.4±455.93μm3 2500.3±229.01μm3,2544.7±359.54μm3, no statistical difference between the groups (P>0.05)Conclusion:seizures do not inevitably cause neuronal loss and not lead to changes of hippocampal volume and neuronal volume in rats with temporal lobe epilepsy, ie seizures do not inevitably cause hippocampal sclerosis.
