Objective:To creat a distress psychological model in pancreatic-cancer-bearing nude mice and explore pancreatic cancer in nude mice growth promoting mechanism of stress.Methods:(1)Creat a distress psychological model in pancreatic-cancer-bearing nude mice. The subcutaneous pancreatic cancer xenografts were established by implanting panc-1 into the back of nude mice. The twenty model mice were evently randomized to two groups:pure-cancer-bearing group(PCB) and PCB plus stress(PCBS) group (n=10/group). The mice in the PCB were supplied with food and water freely. The PCBS was established by putting each nude mouse respectively into a homemade tube. The mice could not move freely in the tubes. Eight hours later, the mice were released from the tubes and then supplied with water and food. After two weeks, mice were sacrificed and blood were collected. (2)The serum epinephrine(E) level was detected with enzyme-linked immunosorbent assay(ELISA). (3)Immunohistochemistry was used to detect the expression of extracellular matrix metalloproteinase inducer(EMMPRIN), Ki-67, vascular endothelial growth factor(VEGF) and CD 105 in transplantation tumors.Results:(1)The weight and volume of transplantation tumors, and serum epinephrine in PCBS were higher(t=-2.812--2.486, P<0.05) than that in PCB. (2)The positive expression of EMMPRIN, Ki-67 and VEGF in PCBS were higher (Z=-2.636~-2.033, P<0.05) than that in PCB. The average of MVD in PCBS(23.58±2.93) was higher(t=-2.846, P<0.05) than that in PCB(20.64±1.44). The expression of EMMPRIN was positively correlated to VEGF and Ki-67(r=0.702; 0.534, P<0.05).Conclusion:Possibly through upregulating the expression of EMMPRIN, Stress induced cancer cell proliferation and angiogenesis, promoting the growth of pancreatic cancer in nude mice,...
