Nowadays, with the increase of people's living standard, diabetes has been becoming the third health killer following the cardiovascular disease and the malignant tumor. The pathogeny and nosogenesis of diabetes are not unclear to date, there are no drug to treat diabetes thoroughly. Although the target of synthetic medicines treated diabetes are clear, yet they have obvious side effects. In China, Traditional Chinese medicines have treated diabetes since ancient. Mulberry leaves are the dried leaves of Morus alba L., which are treated for diabetes from the ancient times. In the recent decades, the hypoglycemic activity of mulberry leaves has received widespread attention.This thesis takes the mulberry leaves as a major subject investigated, aiming at the separation process of total alkaloids from mulberry leaves, the potential purification process of derivative of DNJ and the hypoglycemic activity through the systematic research, the main content of the research work are as follows:1. We use the single factor experiment to investigate the separation process of total alkaloids enriched from mulberry leaves, the best experimental condition of the separation process is 65% ethanol solution, extract 120min*2 times, liquid ratio 1:9. The contents of extracted total alkaloids from mulberry leaves was measured, the best yield was 37.55%.2. Poly-isolated materials, such as C-18 reversed silica gel, sephadex LH-20, were used here to obtained two amino acids and proved the existence of DNJ in total alkaloids, we determined the chemical structure of the purified amino acids through MS, H-NMR and C-NMR, they are proline and phenylalanine, and the relationship between DNJ and proline was discussed.3. Three isolates were tested in the inhibition activity of SGLT2 through the NBDG fluorescence method, the crude extract of mulberry leaves, the total alkaloids of mulberry leaves and the purified DNJ, we proved that the enriched total alkaloids from mulberry leaves possessed obvious hypoglycemic activity.
