| Purpose:To explore the effects of50Hz magnetic fields exposure on PKCζ and related proteins for understanding the role of PKCζ in biological effects of ELF-MF. Materials and methods:FL cells were exposed to a50Hz MF at0.4mT for different durations,then western blotting was used to observe the effects of ELF-MF exposure on PKCζ phosphorylation, and some enzyme inhibitors were applied to study the relationship between PKCζ and its upstream and downstream proteins in cultured FL. Results:Exposure to50Hz MF at0.4mT for5min and15min induced PKCζ dephosphorylation significantly (P<0.05), while it was no effect on exposure for30min and60min.PKCζ dephosphorylation induced by MF was inhibited by PDTC,a specific ROS inhibitor pretreatment.Exposure to50Hz MF at0.4mT for5min,15min,30min, and60min didn't change the level of Bcl-2,Bax and phosphorylated p38MAPK in FL cells (P>0.05). The expression of phosphorylated JNK significantly increased (P<0.05) when exposed to50Hz MF at0.4mT for15min, while there were no effects in other groups. JNK phosphorylation induced by ELF-MF was inhibited by PDTC pretreatment. Conclusion:Exposure to50Hz MF for short duration could induce PKCζ dephosphorylation in FL cells,and ROS might mediated this process.50Hz MF exposure has no effect on p38MAPK phosphorylation, but could induce JNK phosphorylation,and ROS might involve this process.Besides,exposure to50Hz MF did not affect the expression of Bcl-2and Bax in FL cells. |
