Objectives:To expore the effect of HO-1on the inflammatory reaction of rat bile duct ischemia and reperfusion injury and expore the influnce of rat bile salt composition in rat bile duct IRI. Provide some theoretical evidence for the prophylaxis and treatment of bile duct IRI.Methods:Healthy male SD rats128were randomly divided into four groups, the Normal Saline control group (NS group), the adenoviral vector control group (Ad group), adenoviral HO-1experimental group (HO-1group), adenovirus the SiRNA experimental group (SiRNA group), each according to time after reperfusion, divided into1hour(lh),24hour (24h),7days(7d) and14days(14d) four time-groups, each group of each time of eight rats. Products70%hepatic ischemia and reperfusion model (Fei X's method). After45min of ischemia, the clamp was released to allow reperfusion of the liver. After lh,24h,7days,14days to collect the ischemic part of the liver tissue and hepatic bile duct, and the reperfusion period bile of1h group. By Light-microscopy to observe the damage of hepatic cells ande bile duct cells, by electron microscopy to observe the ultrastructural changes of portal bile duct cells. Immunohistochemical (SP method) test CD3+CD45R+T lymphocytes, CD163-Kupffer cells of how extensive the bile duct. Results:1.The display of HE dyeing under Light-microscopy:The pathology damage of HO-1group hepatic and bile duct cells are much lighter than other three groups,SiRNA group is the most serious damage. Suzuki's score has a statistical significance (P<0.01). No obvious difference between NS group and Ad group (P>0.05).2. The display of the ultrastructural structure damage of bile duct cells:SiRNA group of bile duct cells apparent mitochondria swelling, Surface fluffy fall off and Mitochondria crest fracture.The damage of HO-1group are much lighter than other three groups.3. No obvious inflammatory cells infiltrating around bile duct on HO-1group, NS group and group Ad have some inflammatory cells infiltration, but not seriously.No obvious difference between two groups. Serious inflammatory cells infiltration around bile duct on SiRNA group, Among them, the expression of CD3+CD45R+T lymphocytes and CD163+Kupffer cells are much more than other groups, the expression on HO-1group is less.4. Detection of the1h phase in the bile bile salts concentration: Compared with control groups and SiRNA group, the bile salts of HO-1group concentration is lower with statistical significance (P<0.05). Bile duct damage degree is also relatively light. Concentration of SiRNA group are highest (P<0.01). Bile duct damage degree is also relatively serious. No obvious difference between NS group and Ad group (P>0.05).Conclusion:1. Through using recombinant adenovirus construct gene transfection techniques to induce HO-1expression, can reduce the bile duct cells hot ischemia-reperfusion injury, inhibit the expression of HO-1make the damage serious.2. HO-1can reduce inflammatory cells infiltrating around bile duct. May be have correlation with inhibiting the creation of Kuffer cells and T lymphocytes.3. HO-1can reduce the bile duct cells damage through reducing the.bile salt concentration...
