Objective:To Observe the phase change of cyclooxygenase-2(CoX-2) and platelet endothelial cell adhesion molecule (CD31) expression in the laser-induced C57BL/6mice choroidal neovascularization (CNV), as well as COX-2inhibitor celecoxib choroidal neovascularization mechanism to further explore the CNV in the pathogenesis and treatment.Methods:45C57BL/6mice were randomly divided into normal control group, the laser photocoagulation group, celecoxib treatment group, laser photocoagulation group with excessive multi-wavelength krypton ion laser photocoagulation induced choroidal neovascularization. Celecoxib group (medication one week before laser photocoagulation, by gavage, the dose for100Mg/kg once a day, until the end of the experiment). Normal control group in the specimens before photocoagulation, photocoagulation group and the celecoxib treatment group at7days after photocoagulation,14days21days28days the specimens. Material injected before pentobarbital injection lml/kg anesthesia, execution, rapid removal of the eye, drawing, embedded in paraffin, HE staining, COX-2and CD31immunohistochemical staining. Observed by light microscopy photocoagulation damage at the retinal and choroidal tissue changes, the use of the Image-ProPlus6.0software the mean optical density (MOD) of COX-2positive staining to be measured, all data using the SPSS12.0for Windows software packets are processed and analyzed statistically.Results:1,the laser photocoagulation group,21days group microvessel density (MVD) was higher than the7days group,14days group, the difference was statistically significant (P<0.05), MVD in14days and28days between groups no significant difference (P>0.05).2, normal control group, respectively, with the photocoagulation group, plug celebrex about the cloth treatment group at different time points in the treatment group seven days fluctuations in the normal control group, no statistically significant difference, P>0.05; were higher than normal control group, P<0.05, the differences were statistically significant, the photocoagulation group and celecoxib treatment group within the group time point comparisons between the differences were statistically significant (P<0.05).4,After photocoagulation7days of COX-2expression was slightly enhanced at14days was significantly enhanced21days was strongly positive expression at28days showed a continued weakening trend, celecoxib cloth group photocoagulation group at the same time point,7days the two groups no significant difference was not statistically significant (P>0.05) in14days21days28days COX-2positive expression than simply reduce the photocoagulation group, the difference was statistically significant (P<0.05).Conclusion:1, the experimental use of the krypton laser was successfully induced in C57BL/6mice choroidal neovascularization model.2, in the laser-induced choroidal neovascularization, local retinal choroid tissue expression of COX-2protein is increased.3,celecoxib can reduce the expression of COX-2in the laser-induced choroidal neovascularization, which to some the extent to reduce the formation of CNV.
