| Objective:To investigate the Anti-bladder tumor effect in vitro and in vivo of the Improved TAT-VP3Fusion Protein. To research the bladder tumor modle established on Nude Mice.Methods:Apoptosis rate of EJ cells were detected by the method of AnnexinV/7-AAD double staining of flow cytometer. Ultrastructural changes of apoptotic cells were observed by transmission electron microscope. EJ cells were implanted into the bladder and the subcutaneous tissue to establish bladder tumor modle. The group of subcutaneous modle were divided into control group and experimental group. Improved TAT-VP3Fusion Protein was injected subcutaneously to the perimeter of tumor. Calculate the inhibition rate of tumor. Analyze the position which the Protein effect on by IHC.Results:After the effect of the Improved TAT-VP3Fusion protein, EJ cells apoptosis rate was as a time-dose dependent relationship. Apoptosis was observed by transmission electron microscope. Establish orthotopic and subcutaneous bladder tumor modle on nude mice successfully. After the effect of Improved TAT-VP3Fusion Protein, the tumor inhibition rate was38.03%. The result of IHC showed the Protein effect on the nucleus.Conclusion:Improved TAT-apoptin fusion protein in a certain can inhibit the growing of human bladder tumor in vitro and in vivo. Compared the orthotopic and subcutaneous bladder tumor modle on nude mice. We choosed the latter to carry out the animal experiment. |
