Objectives:Nasopharyngeal carcinoma(NPC) was a kind of cancer which was mainly treated by radiotherapy. Due to the different patients have different radiosentivity, so probe the predietors of radiosensitivity in selecting an individual radiotherapy for patients with Nasopharyngeal carcinoma have an important significance. This study was to investigate the Correlation between expression of Cycin D1&Survivin and radiosensitivity of Nasopharyngeal carcinoma, and to find out prediction index for radiosensitivity of Nasopharyngeal carcinoma.Methods:80cases of Nasopharyngeal carcinoma tissue specimens(40cases of radio-sensitive specimens,40cases of radio-resistant specimens), which were obtained before radiotherapy, were selected for HE staining to determine the pathological types; immunohistochemical staining(S-P) detected the expression of Cyclin Dl protein&Survivin protein; Real-time RT-PCR detected the expression of Survivin mRNA and Cyclin D1mRNA; TUNEL detected apoptotic index (AI) of tissue specimens. All statistical data was analyzed with IBM SPSS Statistics20software, usingχ2tes, One-way ANOVA and Spearman rank correlation analysis.Results:(1)The statistical analysis showed that radiosensitivity of Nasopharyngeal carcinoma had a significant relation to histological type, differentiation type and. clinical stage(P<0.05), but it had no significant relation to sex and age(P>0.05).(2)IHC discovered that the high expression rate of Cyclin D1in radio-sensitive group and radio-resistant group were22.5%(9/40) and70%(28/40), which had a significant difference(P<0.05); The high expression rate of Survivin in radio-sensitive group and radio-resistant group were35%(14/40)and77.5%(31/40), which had a significant difference (P<0.05). The expression of Cyclin D1had positive correlation with the expression of Survivin, r=0.313(P<0.05). The expression of Cyclin D1&Survivin had a significant relation to histological type, differentiation type and clinical stage(P<0.05), but it had no significant relation to sex and age(P>0.05).(3)The RQ value of Cyclin D1mRNA in radio-sensitive group and radio-resistant group was16.62±2.24and31.44±2.97, which had a significant difference(P<0.05). The RQ value of Survivin mRNA in radio-sensitive group and radio-resistant group was29.48±3.19and45.38±4.57, which had a significant difference(P<0.05).(4)TUNEL revealed that The AI in radio-sensitive group and radio-resistant group was15.93±2.44and8.56±1.81, which had a significant difference(P<0.05). The AI in high Cyclin D1group and low Cyclin D1group was10.17±2.23and14.2±2.95, which had a significant difference(P<0.05). The AI in high Survivin group and low Survivin group was7.81±1.54and16.68±2.76, which had a significant difference(P<0.05).Conclusion:(1)Radiosensitivity of Nasopharyngeal carcinoma had a significant relation to histological type, differentiation type and clinical stage, but it had no significant relation to sex and age.(2)The expression of Cyclin D1&Survivin had negative correlation with radiosensitivity of Nasopharyngeal carcinoma, which could be a useful predictor of radiosensitivity in NPC.The expression of Cyclin D1&Survivin had a significant relation to histological type, differentiation type and clinical stage, but it had no significant relation to sex and age.(3) Apoptosis had positive correlation with radiosensitivity of Nasopharyngeal carcinoma, it could be a useful predictor of radiosensitivity in NPC. The expression of Cyclin D1&Survivin and apoptosis had negative correlation in NP, it could be a useful predictor of apoptosis.(4)The expression of Cyclin D1and Survivin had positive correlation in NPC. To detect the expression of these two genes can more accurately predict the radiosensitivity of Nasopharyngeal carcinoma.
