| Objective To explore the expression and significance of Matrix Metalloproteinase-12in aortic Dissection.Methods1.Select12cases of the wall of the aorta organization from adult acute aortic dissection Stanford type A patients after surgery as the experimental group,4cases of adult myocardial infarction without aorta wall damage as control group, by reverse transcription polymerase chain reaction, Western Blot, Gelatin Zymography, immuno-histochemistry, fluorescence enzymology and enzyme-linked immuno sorbent assay, analyze and compare the expression of Matrix Metalloproteinase-12between the experimental and control group.2. Select15cases serums of adult acute aortic dissection Stanford type A patients as the experimental group,10cases normal serums as control group. Use fluorescence enzymology and enzyme-linked immuno sorbent assay to analyze and compare the expression of Matrix Metalloproteinase-12between the experimental and control group.Results1. MMP-12in the aorta walls is expressed in both groups, and the expression of the quantity is higher than those in the control group (P<0.05).2. The expression of MMP-12exist in both serums. The expression of the experimental group is obviously higher than those in the control group (P<0.05).3. Expression of MMP-12in the aorta wall is higher than serum in aortic dissection(P<0.05).4. MMP-12of the aorta wall and serum all can be inhibited by MPIs (P<0.05).Conclusion1. The expression of MMP-12in aorta wall of acute aortic dissection Stanford type A is higher, which shows that MMP-12participated in the formation of acute aortic dissection Stanford type A and the remodeling processes of aorta walls.2. The expression of MMP-12in serums of acute aortic dissection Stanford type A is significant higher than normal group, which shows that MMP-12and acute aortic dissection Stanford type A is closely related to the occurrence and plays an important role in its development.3. The expression of MMP-12in aorta wall of aortic dissection is higher than that in serum, which shows that MMP-12accumulated in the intima broken and release into the blood; MMP-12participated in the aorta wall damage process and plays a very important role in the process of aortic dissection formation and development.4. MMP-12can be inhibited by MPIs, which is very vital to the further study of MMP-12specific inhibitors on preventing the damage of aorta wall. |
