Objective: Lung cancer is the most common malignant tumors ofthorax,acount for80%of non-small cell lung cancer, Its characteristic isaggressive, and a high death rate,the most of patients had distant metastasesoccurred before they be pathological diagnosed,lost the best time for surgicaltreatment. Metastasis and recurrence are the the bottleneck of survival rates ofnon-small cell lung cancer, The first step that malignant tumor cells realizationof invasion and metastasisis breach the barrier which fromed by BM andECM.Heparanase is an endo-β-glucuronidase that specifically cleavescarbohydrate chains of heparan sulphate proteoglycans (HSPG) which befromde by core protein and several heparan sulfat eproteoglyean(HSPG) withno covalent bond, HSPG is not only the formation of extracellularmicroenvironment is main form, but also the natural substrates of heparanase.In malignant tumor molecular biology study found that Histonedeacetylase6(HDAC6) is microtubule and HSP90associated protein whichregulation of gene transcription by microtubules to acetylation and HSP90deacetylation, destructioning the " environment" steady of vivo acetylationand breaking the "dynamic balance" of histone acetylation and deacetylation.HDAC6involved in tumor metastasis and tumor angiogenesis by increasingthe athletic ability of tumor cells and Vascular endothelialMethods: We collected56cases Clinical Hospital of Hebei MedicalUniversity Fourth in the Department of thoracic,which Surgically resected andhistologically confirmed NSCLC. Before the coming surgery therapy, all thepatients received neither radiotherapy nor chemotherapy. Information aboutclinicopathological characteristics, such as sex and age of the patients, lungcancer cell differentiation and histological type, lymph node metastasis,(TNM)stage, Detect the expressions of HPA and HDAC6in56tumor specimens and in22normal lung tissue (Collected by operation resection of pulmonarybenign lesions of the lung tissue specimens from22cases which wereconfirmed by pathology (after operation for benign lesions, IncludingPulmonary bulla3cases; Hamartoma5cases; Bronchiectasis2cases; Lungabscess1cases; Inflammatory pseudotumor2cases;Pulmonarymycosis1cases)by Immunohistochemistry.χ~2test and Spearman Correlation in SPSS forwindows13.0are used in the study.Results:1. Immunohistochemistry was employed to detect the expression of HPAprotein in56specimens of non-small cell lung cancer tissue and22specimensof normal lung tissue. The positive expression rate of HPA protein in non-small cell lung cancer tissues was55.4%(31/56), The normal lung tissues was13.6%(3/22). There was significant difference in the positive rates of HPAprotein between non-small cell lung cancer tissues and the normal lung tissues(χ~2=11.181,P=0.001<0.05). Expression of HPA wasn't correlated withpatients' gender, age, histological type and differentiation (P>0.05), but it wascorrelated with lymph node metastasis, TNM stage (P<0.05).2. Immunohistochemistry was employed to detect the expression ofHDAC6protein in56specimens of non-small cell lung cancer tissue and22specimens of normal lung tissue. The positive expression rate of HDAC6protein in non-small cell lung cancer tissues was64.3%(36/56), The normallung tissues was18.2%(4/22). There was significant difference in the positiverates of HDAC6protein between non-small cell lung cancer tissues and thenormal lung tissues (χ~2=13.438,P=0<0.05). Expression of HDAC6wasn'tcorrelated with patients gender, age, differentiation and histological type and(P>0.05), but it was correlated with lymph node metastasis and TNM stage(P<0.05).3. The HPA positive expression rat in the tissue of the HDAC6-positivewas69.4%,There was a positive correlation between HDAC6and HPA in non-small cell lung cancer tissues (r=0.385P=0.003<0.05) Conclusion: The over-expression of HPA HDAC6in NSCLC wasn'tcorrelated with patients' gender, age, histological type and differentiation,but itwas correlated with lymph node metastasis, TNM stage. The expression ofHPA HDAC6with TNM staging is increasing,The expression of HPAandHDAC6in NSCLC with lympt node metastasis is higher than those withoutlympt node metastasis, HPA HDAC6may play an important role in NSCLCmetastasis,The expression of HPA had positive correlation with HDAC6inNSCLC.The over-expression of HPA HDAC6may be an inportant factor inthe development of NSCLC.It maybe a index that assessed the invasion andprognosis of lung cancer.
