Objective:The extraction and identification of purple sweet potato anthocyanin; This study is to investigate the crude extract of purple sweet potato anthocyanin on antioxidant and lipid metabolism in vivo as well as about the regulatory role on acetyl coenzyme A carboxylase gene (ACC1).Methods:(1) With the way of ultrasonic extraction and freeze-dissolve to purify the purple sweet potato anthocyanin, and the content were determined by UV spectrophotometer.(2)80male mice(8w,KM) were divided into the health group and high lipid group randomly. Among them, the health group is split into four groups (A/B/C/D), which were gavaged anthocyanin as follow:0,50,100,200mg/kg of bw, fed the basal diet. High-lipid group is also divided into four groups (E/F/G/H), the same intragastric dose as above, fed with high fat diet. This test last30d. the serum were determination of total cholesterol (TC), triglyceride (TG), high-density lipoprotein (HDL) and low density lipoprotein (LDL) level. Collected liver, weighing the analysis of liver index and histopathological examination of the liver,the slurry is used to detect superoxide dismutase (SOD), lipid peroxidation (MDA) and glutathione peroxidase (GSH-PX),.(3) Detection of anthocyanin regulation of acetyl-CoA carboxylase gene (ACC1) in the L02cell with RT-PCR.Results:(1) Comparison of the two purification methods, showing that the method of ultrasonic extraction and freeze-dissolve is better, extract purity of purple sweet potato anthocyanin is18.9%, and the content is5mg/g dry powder.(2) mice weight results show that the mice's weight of high-lipid group decreased significantly (P<0.05). Liver coefficient suggest that mice's liver index decreased significantly in the health group (P<0.05); there is no significant difference in the group of high-fat group. The health group mice liver without significant pathological changes. There is a slight enlargement of the liver in high-fat group, partial color khaki, dim texture toughness, fragile. The biopsy showed that high-fat mice liver cells with varying degrees of vesicular degeneration and fatty degeneration. The serum test results show that the level of TC, TG and LDL decreased in varying degrees, and increaseing HDL in the health group; compared with the control group, the level of TC and HDL have increased, TG and LDL have decreased in the lipid group. In the slurry of the liver tissue, both of the health group and high-fat mice show that the activity of SOD and GSH-PX have increased (P<0.05), and MDA have decline (P<0.05).(3) RT-PCR results show that compared with the control group, the ACC1expression with low dose of anthocyanins in the lipid model cells were significantly increased.Conclusion:(1) Compared with the heating method, ultrasonic extraction-freeze to dissolve is more efficient to the extraction of purple sweet potato anthocyanin.(2)the effect on lossing weight were significant in high-lipid model with purple potato anthocyanins;(3) the effect on antioxidant and the lipids regulation of purple sweet potato anthocyanin were significant;(4) In the high lipid cell model, middle and high concentrations of anthocyanins (2.5ug/mL,20ug/mL) can reduce the expression of ACC1, and result in reducing synthesis of fatty acid.
