Objective: In order to discuss the effect and influence of Achilles tendon in rats by the Low level laser therapy (LLLT), the experiment used male Wistar rats as research objects, and observed the contents of hydroxyproline and gene expression of Achilles tendon Collagen-I and TGF-β1 by LLLT after exercising by histology, biochemistry and molecular biology techniques.Methods: The experiment adopted two kinds of model - jumping by the electricity and LLLT: 96 male Wistar rats were selected randomly as the control group (n=8) and the exercise group (n=88), the rats of exercise group were divided randomly into two groups again:①The exercise control group (n=8)②Nature healing group (n=40). According to the time of samples been taken, the groups were further allocated to 1, 2, 3, 7, 14d after exercising. Each group has 8 rats.③LLLT group (n=40). LLLT groups were also allocated to 1, 2, 3, 7, 14d after exercising. Each group has 8 rats. The rats of exercise were trained for 5 weeks, and then we use the LLLT on the rats of LLLT groups depending on the different needs. Text methods and Index:①Observing the vary of the tendon collagen in rats.②Samples alkaline hydrolysis measures the contents of hydroxyproline of the tendon.③The expression of Collagen-I and TGF-β1 in mRNA level were analyzed by real-time quantitative PCR technique.Results:①During 3d of LLLT, LLLT group's contents of hydroxyproline and the gene expression of Collagen-I and TGF-β1 have no significant difference with the exercise control group's (p>0.05) and nature healing group's (p>0.05) according to the time of samples been taken.②On the 7th day, contract with the exercise control group, LLLT group's gene expression of Collagen-I are increased significantly (p<0.05) ,contents of hydroxyproline and the gene expression of TGF-β1 have no significant difference (p>0.05). LLLT group's gene expression of Collagen-I and TGF-β1 are higher significantly than nature healing group's (p<0.05) according to the time of samples been taken.③On the 14th day, contract with the exercise control group, LLLT group's contents of hydroxyproline and the gene expression of Collagen-I are increased significantly (p<0.01), the transcription of TGF-β1 varys no difference (p>0.05). LLLT group's contents of hydroxyproline (p<0.05) and the gene expression of Collagen-I and TGF-β1 (p<0.01) have very significant difference with Nature healing group's.Conclusion:①Low level laser has some effects on Achilles tendinopathy enthesiopathy in rats in early stage. This starts from 7d and affects significantly in 14d.②Low level laser can improve the anabolic collagen and contents of hydroxyproline in transcription level in Achilles tendinopathy enthesiopathy in rats. This indicates Low level laser can improve the anabolic collagen and promote the collagen metabolism.③Low level laser can delay the down-regulation of the expression of TGF-β1 in transcription level. It can keep the TGF-β1 in the high level between 7d and 14d. Low level laser plays a role part in regulating the extracellular matrix and accelerate the speed of Achilles tendon healing in rats in early stage.
