Objective1.To explore the association between polymorphism of peroxisomeproliferator-activated receptors(PPARs)single nucleotide polymorphisms (SNPs)and serum C-reactive protein (CRP) levels.2. To investigate the association between PPAR α/δ/γ haplotypes and serumC-reactive protein (CRP) levels.3. To discuss whether the interaction of PPARs haplotypes with obesity affectsCRP level.MethodsSubjects (N=644) were randomly selected from Prevention of Multiplemetabolic disorders and Metabolic syndrome in Jiangsu province (PMMJS). TenSNPs (rs135539, rs1800206, rs4253778, rs2016520, rs9794, rs10865710,rs1805192, rs4684847, rs709158, rs3856806) of PPARs were selected. Polymerasechain reaction restriction fragment length polymorphism was used to detect thepolymorphisms of rs4253778while Taqman fluorescence probe method was usedin other SNPs. Variance test and t test were used to analyze association betweenPPARs genotypes and CRP level. The association between PPARSs haplotypes andserum CRP levels and the interaction of PPARs haplotypes with obesity wereanalyzed using SNPStats software.Results1. Minor alleles of rs135539, rs1800206and rs9794were associated withchanges in CRP level (P <0.05). The rs135539-C allele was associated withincreased CRP level and rs1800206-V, rs979-G allele with decreased CRP level.2. After adjusting for sex, age, blood pressure, cigarette smoking, alcoholdrinking, haplotypes of AVG and CVG in PPARα, CG in PPARδ were associated with increased CRP level (P <0.05) while haplotypes of CC in PPARδ, CPCAC inPPARγ were associated with decreased CRP level (P <0.05).3. Haplotype-obesity interaction analysis also noted interaction of obesity withAVG and CVG in PPARα and CG in PPARδ.Conclusions1.3SNPs of PPARs are associated with increased or decreased CRP level.2. Haplotypes of AVG, CVG, CG, CC and CPCAC in PPARs are associated withincreased or decreased CRP level.3. Haplotypes of AVG, CVG and CG in PPARs interact with obesity to set CRPlevel.
